Summary: This study characterizes the transcriptome and translatome of the arterivirus porcine reproductive and respiratory syndrome virus (PRRSV), and analyzes the host response to infection. The study reveals an increase in programmed ribosomal frameshift (PRF) efficiency over time at both viral genomic sites. Surprisingly, the study also finds an increase in PRF efficiency at the canonical ORF1ab frameshift site, contradicting the common assumption. Additionally, several highly translated additional viral ORFs and novel viral transcripts were discovered. These findings provide insights into the gene expression programs of PRRSV.

This study revealed that the L1C-1-4-4 variant evolved from a recombinant ancestor between the L1C and L1A genes. The emergence of this variant and its rapid spread highlight the role of inter-sub-lineage recombination in the evolution of PRRSV-2.

Summary: The ZMAC cell line is more advantageous for isolating PRRSV from lung and serum samples compared to the MARC-145 cell line. However, not all PRRSV isolates obtained from ZMAC cells are able to grow in MARC-145 cells. This report provides important guidelines to improve PRRSV isolation from clinical samples for further characterization and/or production of autogenous vaccines.

Emergence of a New Lineage 1C Variant of Porcine Reproductive and Respiratory Syndrome Virus 2 in the United States
