PARP inhibition impedes the maturation of nascent DNA strands during DNA replication

Alina Vaitsiankova et al.

Summary: PARP inhibitors hinder the maturation of nascent DNA strands during DNA replication, particularly unligated Okazaki fragments and other discontinuities, resulting in cytotoxic effects. The activation of PARP1 is elevated in cells lacking the FEN1 nuclease, suggesting its involvement in the detection and processing of these DNA replication intermediates. PARP inhibitors disrupt the integrity of nascent DNA strands in both normal and FEN1(-/-) cells, leading to the formation of single-strand nicks or gaps. These findings highlight the importance of unligated Okazaki fragments and other discontinuities in the cytotoxicity of PARP inhibitors.

Limiting DNA polymerase delta alters replication dynamics and leads to a dependence on checkpoint activation and recombination-mediated DNA repair

Natasha C. Koussa et al.

Summary: Pol delta is crucial for eukaryotic DNA replication, and depletion of Pol delta leads to slower replication progression and genome instability. Impaired lagging-strand synthesis due to Pol delta limitation results in increased single-stranded DNA and DNA damage, requiring a strict dependence on checkpoint signaling and repair pathways.

HPF1-dependent PARP activation promotes LIG3-XRCC1-mediated backup pathway of Okazaki fragment ligation

Soichiro Kumamoto et al.

Summary: Studies have shown the essential role of PARP1-HPF1 in LIG3-dependent Okazaki fragment joining, serving as a backup system for when LIG1 is nonfunctional. In the absence of LIG1, LIG3-XRCC1 can be recruited to chromatin for Okazaki fragment joining. Depletion of PARP1 or HPF1 leads to failure in recruiting LIG3 to chromatin, while co-depletion of LIG1 and XRCC1 completely prevents Okazaki fragment joining.

Daughter-strand gaps in DNA replication - substrates of lesion processing and initiators of distress signalling

Ronald P. Wong et al.

Summary: Dealing with DNA lesions during genome replication is challenging, but repairing daughter-strand gaps through translesion synthesis or recombination-mediated template switching is crucial for genome maintenance. Proper control over the activities of daughter-strand gaps has important implications for cancer therapy.

Replication gaps are a key determinant of PARP inhibitor synthetic lethality with BRCA deficiency

Ke Cong et al.

Summary: Mutations in BRCA1 or BRCA2 are found to be synthetic lethal with PARPi, with toxicity deriving from replication gaps instead of DNA double-stranded breaks. The study uncouples HR, FP, and fork speed from PARPi response, highlighting gaps as a determinant of PARPi toxicity and changing the paradigm for synthetic lethal interactions.
