A NIR fluorescent probe for fatty acid amide hydrolase bioimaging and its application in development of inhibitors

Summary: A highly selective and sensitive FAAH-activated near-infrared fluorescent probe named DAND was developed for real-time detection and imaging of FAAH activity, leading to the identification of piperine as a novel inhibitor of FAAH. Based on the interaction with piperine, a more potent FAAH inhibitor (11f) with excellent anti-inflammatory activity was designed and synthesized. These findings suggest that DAND can be a promising molecular tool for exploring FAAH activity and for screening potential FAAH inhibitors, while piperine and its derivatives may serve as potential candidate drugs for the treatment of neurodegenerative diseases in the future.
