A case study on branched aliphatic carboxylic acids shows the potential to induce hepatic steatosis. By analyzing gene expression patterns and adverse outcome pathways, researchers were able to confirm biological similarity and design an in vitro testing battery to systematically investigate a common mode of action among the compounds.

A tiered approach to population-based in vitro testing for cardiotoxicity: Balancing estimates of potency and variability

Summary: Population-wide in vitro studies using human induced pluripotent stem cell-derived cardiomyocytes (hiPSC-CM) are effective in assessing cardiotoxicity hazard and population variability. Computational down-sampling analysis can optimize study design and determine the appropriate population size considerations. The study found that a cohort of 5 unique donors is sufficient for accurate estimation of potency and QT prolongation risk, while a cohort of at least 20 donors is needed for estimating inter-individual variability.

However, it is still in its early stages and further case studies are needed to determine if safety decisions are sufficiently protective and not overly conservative. High-Throughput Transcriptomics Platform for Screening Environmental Chemicals

Summary: This study evaluated the TempO-Seq assay for HTTr concentration-response screening of chemicals in the human-derived MCF7 cell model. A robust bioinformatics pipeline was developed using open-source tools, along with a novel gene expression signature-based concentration-response modeling approach. Analysis showed highly reproducible differential gene expression signatures, along with in vitro transcriptional biological pathway altering concentrations closely aligned with previous high-throughput screening assays.
