A review on age-related cancer risks in PTEN hamartoma tumor syndrome

Linda A. J. Hendricks et al.

Summary: Patients with PTEN hamartoma tumor syndrome have an increased risk of cancer due to pathogenic PTEN germline variants. Studies have summarized the specific cancer risks for these patients, providing valuable guidance for clinical management. However, caution should be exercised when interpreting the data due to discrepancies between studies.

Natural History of Thyroid Disease in Children with PTEN Hamartoma Tumor Syndrome

Jessica R. Smith et al.

Summary: Thyroid disease is common in children with PHTS, with females being diagnosed with nodules earlier than males. Female sex and the absence of neurological findings of PHTS are significantly associated with the risk of thyroid nodules. Abnormal lymph nodes with echogenic foci were observed by ultrasound in 20% of patients, but were not linked to malignancy. Autoimmune thyroid disease was present in 30.3% of assessed subjects.

The Clinical Spectrum of PTEN Hamartoma Tumor Syndrome: Exploring the Value of Thyroid Surveillance

Julia A. Baran et al.

Summary: PHTS patients commonly present with macrocephaly, impaired development, skin/oral lesions, and autism spectrum disorder. Initiating thyroid ultrasound surveillance before the age of 10 does not appear to confer a clinical advantage, with thyroid nodules in patients over 10 more likely to be benign.

Thyroid nodular disease and PTEN mutation in a multicentre series of children with PTEN hamartoma tumor syndrome (PHTS)

Gerdi Tuli et al.

Summary: In a multicentre study of pediatric patients with PTEN mutations, the incidence and characteristics of nodular thyroid disease were reported, with no evidence of malignancy. Correlations were found between unrelated factors such as head circumference and multiple lipomas with thyroid lesions.
