Summary: This study evaluated the use of reversible inhibition of DNA replication to reduce mosaicism in embryos edited by CRISPR/Cas9. The findings showed a concentration-dependent DNA replication inhibition, leading to a decrease in mosaicism but a detrimental effect on embryo development. Balancing the use of this approach is necessary considering the impact on embryo development.

Summary: This study successfully generated CD163-modified pigs by introducing the CD163 gene into porcine zygotes using the CRISPR/Cas9 system. Three out of the four tested gRNAs showed high editing efficiency, but further improvements are needed for generating genetically modified pigs with high efficiency.

One-Step Generation of Multiple Gene-Edited Pigs by Electroporation of the CRISPR/Cas9 System into Zygotes to Reduce Xenoantigen Biosynthesis

Summary: The study successfully utilized a CRISPR/Cas9 system to simultaneously target GGTA1, CMAH, and B4GALNT2 in in vitro-fertilized zygotes, achieving the one-step generation of multiple gene-edited pigs without xenoantigens. Although the experiments resulted in the generation of GGTA1/CMAH double-edited pigs and GGTA1/CMAH/B4GALNT2 triple-edited pigs, these gene-edited pigs were genetic mosaics, indicating further refinement is needed to completely knock out all xenoantigens.

This study found that the gene editing method and embryonic stage for gene editing can significantly impact the gene editing efficiency and mutation rates of porcine embryos.
