The Ts-group on the ynamide serves as a directing group, leading to the formation of alkenyl nickel species with high regioselectivity.

Merging NiH Catalysis and Inner-Sphere Metal-Nitrenoid Transfer for Hydroamidation of Alkynes

Summary: A NiH-catalyzed strategy for the hydroamidation of alkynes with dioxazolones has been reported in this study, providing synthetically useful secondary enamides in (E)-anti-Markovnikov or Markovnikov selectivity. The reaction is viable for various alkynes and functional groups, with H2O found to be essential for high catalyst turnovers. Inner-sphere nitrenoid transfer is proposed as a key component in outcompeting undesired processes in the Ni catalysis.

Facile Synthesis of Chiral Arylamines, Alkylamines and Amides by Enantioselective NiH-Catalyzed Hydroamination

Summary: Regio- and enantioselective hydroarylamination, hydroalkylamination and hydroamidation of styrenes have been developed using NiH catalysis with a simple bioxazoline ligand under mild conditions. A variety of enantioenriched benzylic arylamines, alkylamines and amides can be easily accessed by employing nitroarenes, hydroxylamines and dioxazolones as amination reagents. Chiral induction in these reactions is proposed to proceed through an enantiodifferentiating syn-hydronickellation step.

Mechanistic Investigation and Optimization of Photoredox Anti-Markovnikov Hydroamination

Summary: The selectivity and reaction mechanism for the photocatalytic anti-Markovnikov hydroamination of unactivated alkenes with primary amines to produce secondary amines has been revealed through time-resolved laser kinetics measurements of key reaction intermediates. The absence of back-electron transfer between the photogenerated aminium radical cation and reduced photocatalyst complex leads to the selectivity for primary amine alkylation. The turnover of the photocatalyst occurs through a reaction between Ir(II) and a thiyl radical, with the in situ formation of an off-cycle disulfide suppressing this turnover and reducing reaction efficiency.

NiH-Catalyzed Hydroamination/Cyclization Cascade: Rapid Access to Quinolines

Summary: A highly efficient NiH catalytic system was developed for the selective hydroamination of alkynes, leading to structurally diverse quinolines. This method is applicable to a wide range of alkynes and has shown utility in the functionalization of natural products and the synthesis of complex molecules. Preliminary mechanistic experiments suggest an alkenylnickel-mediated alkyne hydroamination and intramolecular cyclization of enamine intermediates.
