Local inhibition of TGF-β1 signaling improves Th17/Treg balance but not joint pathology during experimental arthritis

Summary: TGF-beta 1 is a crucial growth factor for promoting the differentiation of Th17 and Treg cells. Inhibition of TGF-beta 1 with the inhibitor SB-505124 effectively suppresses Th17 differentiation and improves the Th17/Treg balance. However, local treatment with SB-505124 does not inhibit joint inflammation and destruction in an in vivo experimental arthritis model.

Antigen-presenting innate lymphoid cells orchestrate neuroinflammation

Summary: This study identified an inflammatory population of group 3 innate lymphoid cells (ILC3s) that play a critical role in a mouse model of multiple sclerosis. These ILC3s stimulate myelin-specific T cells through antigen presentation, leading to T cell responses in the CNS and disease development.

Summary: The study found significantly increased relative frequencies of ILC2 and ILC3 in decidua and an increase of ILC2 in cord blood samples in the preterm birth group compared to full-term birth samples. Additionally, there was a decrease in IFN-gamma in peripheral blood samples of the preterm birth group, suggesting functional withdrawal. Levels of IL-4, IL-17, IL-22 were similar in preterm birth and full-term birth groups, indicating a relevant role in mediating labor.

Summary: Innate lymphoid cells (ILCs) are the most abundant immune cells in the uterine mucosa before and during pregnancy, with tissue-resident natural killer (NK) cells and ILCs being the main subsets. Recent advances in single-cell RNA sequencing and high dimensional flow and mass cytometry have significantly enhanced our understanding of the heterogeneity within the uILC compartment. These advancements may help answer key clinical questions regarding infertility, miscarriage, and pregnancy disorders related to uILC subsets.
