Severe de novo pustular psoriasiform immune-related adverse event associated with nivolumab treatment for metastatic esophageal adenocarcinoma

Riyad N. H. Seervai et al.

Summary: Breakthrough targeted therapies have greatly improved survival for cancer patients, but can lead to cutaneous immune-related adverse events (irAEs). Psoriasiform irAEs, while rare, can be severe and may require treatment with biologic therapy based on clinical and histopathologic examinations to identify patients who may benefit.

Interferon-Gamma-Producing CD8+ Tissue Resident Memory T Cells Are a Targetable Hallmark of Immune Checkpoint Inhibitor-Colitis

Sarah C. Sasson et al.

Summary: Our study demonstrates that CD8(+) tissue resident memory T (T-RM) cells are the main activated T cell subset in ICI colitis. The pattern of gastrointestinal immunopathology in ICI colitis is distinct from ulcerative colitis at both the immune and epithelial-signaling levels. Activation of CD8+ T-RM cells correlates with clinical and endoscopic severity of ICI colitis.

Summary: Late-onset and long-lasting immune-related adverse events (irAEs) during treatment with ICIs are common but underreported events that require time-dependent survival analysis to evaluate their impact on overall survival.

Expansible residence decentralizes immune homeostasis

Sathi Wijeyesinghe et al.

Summary: The immune system in adult mice is integrated within tissues, with a durable maintenance of resident memory T cells contributing to tissue immunity. Tissue pliancy facilitates the accumulation of tissue-resident memory without eroding pre-existing antiviral T cell immunity, resulting in the decentralized homeostasis of organismal immunity.

Chronic Immune-Related Adverse Events Following Adjuvant Anti-PD-1 Therapy for High-risk Resected Melanoma

J. Randall Patrinely et al.

Summary: This multicenter cohort study found that chronic immune-related adverse events (irAEs) associated with anti-PD-1 therapy were more common than previously recognized, with a considerable proportion of patients developing chronic irAEs such as endocrinopathies and arthritis. Visceral organ-related irAEs had lower rates of becoming chronic, highlighting the importance of considering the risks of chronic irAEs in treatment decision-making.

Endogenous retroviruses promote homeostatic and inflammatory responses to the microbiota

Djalma S. Lima-Junior et al.

Summary: The skin microbiota stimulates the expression of endogenous retroviruses (ERVs), leading to the induction of commensal-specific T cells and immune responses. Inhibition of ERV reverse transcription impairs immunity and tissue repair, while a lipid-enriched diet enhances ERV expression for heightened immune responses and inflammation in response to colonization by commensal microbes.

Jacobi Hines et al.

Summary: The patient developed two high-grade unusual irAEs despite receiving pembrolizumab treatment and B cell-directed immunosuppressive therapy. His symptoms were ultimately improved with tacrolimus.
