Influence of surface interaction between drug and excipient in binary mixture for dry powder inhaler applications

Neetu Varun et al.

Summary: This study investigates the drug-excipient incompatibility in physical mixtures and its impact on properties of dry powder inhalers. The findings reveal the formation of condensed or addition compound due to interaction between drug and excipient in solid-state. The study also suggests that surface modification of excipient can improve the performance of the mixtures.

Crystallization induced flower-like lactose as potential carriers for dry powder inhaler application

Neetu Varun et al.

Summary: This study highlights the use of flower-like lactose particles as potential carriers to improve the efficiency of Dry Powder Inhalers (DPIs). The results show that these lactose particles have higher surface roughness, amorphicity, and surface energy compared to commercial lactose carriers, leading to higher fine particle fraction (%FPF) and improved inhalation efficiency.

Design and evaluation of inhalable nanocrystals embedded microparticles with enhanced redispersibility and bioavailability for breviscapine

Yingchong Chen et al.

Summary: A novel inhalable nanocrystals-embedded microparticles system for pulmonary delivery of breviscapine (BVC) was designed in this study. The formulation, including TPGS-modified BVC nanocrystals and BVC-NEP, demonstrated significant influence of matrix formers type and concentration on particle morphology, drug release, and lung deposition. The optimized formulation with TPGS as stabilizer and MN as matrix formers showed the highest FPF value and excellent pharmacokinetic performance.

Spherical agglomerates of lactose as potential carriers for inhalation

Sarah Zellnitz et al.

Summary: This study reveals that spherical lactose agglomerates have potential as carriers for inhalation applications. Compared to a standard lactose inhalation grade carrier, the spherical lactose agglomerates have larger specific surface area and higher fine fraction, which enhances the dose of active pharmaceutical ingredient and improves aerosolization performance. The carrier material attributes related to particle size, specific surface area, compressibility, flowability, and air permeability are critical for aerosolization performance.

Ultrasound-assisted solution crystallization of fotagliptin benzoate: Process intensification and crystal product optimization

Lan Fang et al.

Summary: The ultrasound-assisted crystallization process showed potential in improving process efficiency and modifying crystalline product properties for fotagliptin benzoate methanol solvate (FBMS). It was found that ultrasound could effectively reduce crystal size, narrow the metastable zone width, and shorten the induction time, leading to enhanced desolvation kinetics during the drying process. Small rod-shaped crystals produced by ultrasound irradiation exhibited faster desolvation compared to long needle crystals obtained by conventional crystallization processes.
