Phosphorylation of the alpha-subunit of plant eukaryotic initiation factor 2 prevents its association with polysomes but does not considerably suppress protein synthesis

Andrey Zhigailov et al.

Summary: The phosphorylation of plant eIF2 does not greatly affect its ability to participate in the initiation of mRNA translation, in contrast to animals and yeast, in which eIF2 alpha phosphorylation results in profound suppression of protein synthesis. This was shown through experiments in tobacco and wheat, where phosphorylated plant eIF2 was still able to participate in translation initiation without significant reduction in polysome abundance.

Ribosome quality control antagonizes the activation of the integrated stress response on colliding ribosomes

Liewei L. Yan et al.

Summary: Translation stalling activates ribosome quality control (RQC) and is linked to the integrated stress response (ISR) through Gcn2, with Hel2 suppressing Gcn2 activation. Both pathways are activated by similar factors causing ribosome stalling, although Hel2 is maximally activated at a lower frequency. Inactivation of one pathway leads to overactivation of the other, suggesting they are both triggered by the same signal. ISR prefers an empty A site while RQC shows no preference, indicating they do not directly compete with each other.

Diverse nitrogen signals activate convergent ROP2-TOR signaling in Arabidopsis

Yanlin Liu et al.

Summary: Independent of nitrogen assimilation, nitrate and ammonium act as primary nitrogen signals to activate TOR in the Arabidopsis leaf primordium. Additionally, 15 proteinogenic amino acids can activate TOR, with the first amino acids generated from plant specific nitrogen assimilation, sulfur assimilation, and glycolate cycle showing the highest potency.

Global phosphoproteomics pinpoints uncharted Gcn2-mediated mechanisms of translational control

Ladislav Dokladal et al.

Summary: Gcn2 protein kinase mediates cellular adaptations to amino acid limitation by phosphorylating eIF2 alpha, as well as targeting auxiliary effectors such as eIF2 G protein complex for translational control. This mechanism contributes to the inhibition of translation initiation in amino-acid-starved cells and plays a more intricate role in translation control than previously understood.

Structure of Gcn1 bound to stalled and colliding 80S ribosomes

Agnieszka A. Pochopien et al.

Summary: The Gcn pathway is highly conserved in eukaryotes, with Gcn1 serving as an essential effector protein for Gcn2 kinase to regulate global translation in response to stress. The structure of Gcn1 interacting with stalled and colliding ribosomes was revealed through cryo-electron microscopy, shedding light on the activation of Gcn2. This study provides important insights into the molecular mechanism of Gcn pathway activation and its implications in ribosome-associated quality control pathways.

Plant Stress Granules: Trends and Beyond

Israel Maruri-Lopez et al.

Summary: Stress granules (SGs) are dynamic membrane-less condensates formed through liquid-liquid phase separation (LLPS) in response to stress, with a biphasic structure consisting of core and shell components. The core is composed of proteins with intrinsically disordered regions and RNA-binding domains, while the shell contains species-specific components like metabolic enzymes and transcription factors. SGs play a crucial role in cellular response to stress and post-stress recovery, and their disassembly after stress allows for the reactivation of translation and cell growth.
