Variation in cis-regulation of a NAC transcription factor contributes to tolerance in wheat

Summary: Drought is a major limitation to wheat production worldwide, and understanding the genetic components determining drought tolerance is crucial for wheat breeding. In this study, we identified a wheat NAC gene (TaNAC071-A) that is strongly associated with drought tolerance. Overexpression of TaNAC071-A improved drought tolerance in wheat through increased expression of stress-responsive genes and improved water-use efficiency. We also found that a 108-bp insertion in the promoter region of TaNAC071-A contributes to variation in drought tolerance among wheat accessions. Importantly, introgression of this insertion allele into drought-sensitive cultivars improved their drought tolerance, making it a valuable genetic resource for wheat breeding.

PPVED: A machine learning tool for predicting the effect of single amino acid substitution on protein function in plants

Summary: This study investigated the effects of single amino acid substitutions on protein function in plants and developed a plant SAAS database and a prediction tool called PPVED. The results showed that PPVED had a high predictive accuracy for plant SAASs, surpassing other human database software. Wet lab experiments validated the predictions made by PPVED, demonstrating its reliability in identifying and characterizing genetic variants that explain phenotype variations in plants, and contributing to solutions in functional genomics and systems biology challenges.

Revisiting the role of delta-1-pyrroline-5-carboxylate synthetase in drought-tolerant crop breeding

Summary: The P5CS gene plays a protective role in stressed plants, but overexpression of P5CS may inhibit plant growth. By expressing AtP5CS1 and AtP5CS1F128A genes under the regulation of an ABA-inducible promoter, it was found that seedling growth and grain yield of rice can be increased, but it has detrimental effects on grain quality, tiller number, and grain yield.

A teosinte-derived allele of a MYB transcription repressor confers multiple disease resistance in maize

Summary: This study cloned a teosinte-derived allele of a resistance gene, ZmMM1, in maize, which confers resistance to multiple diseases and acts as a transcription repressor. The mechanism involves polymorphisms in the 3' untranslated region of ZmMM1 leading to increased protein accumulation, ultimately regulating specific target genes involved in plant immunity and cell death. The successful isolation of the ZmMM1 resistance gene provides insights for developing broad-spectrum disease resistance and understanding molecular mechanisms underlying multiple disease resistance.
