Summary: This study used cysteine-scanning mutagenesis to assess the functional effects of Cys-modifying reagents on the palm domain beta 10 strand residues in mouse ENaC. Transmembrane serine protease 2 (TMPRSS2) proteolytically activates the epithelial sodium channel (ENaC) by cleaving the channel's γ-subunit

Summary: The study shows that human TMPRSS2 can stimulate channel activity in heterologously expressed ENaC, likely through increasing channel open probability. Additionally, TMPRSS2 plays a dual role in the release of the gamma-inhibitory tract.

Summary: The regulation mechanism of ENaC phosphorylation is not fully understood. Phosphorylation of serine residue S621 activates ENaC and primes serine residue S617 for subsequent phosphorylation by GSK3 beta resulting in channel inhibition. Additionally, DYRK2 stimulates ENaC currents while GSK3 beta inhibits the currents.

Dissecting the Effects of Aldosterone and Hypokalemia on the Epithelial Na+ Channel and the NaCl Cotransporter

Mathias Kristensen et al.

Summary: Primary hyperaldosteronism (PA) is a condition characterized by excessive aldosterone production and high blood pressure. This study investigates the effects of aldosterone and potassium on sodium channels in the kidneys and suggests that both aldosterone and potassium can directly influence sodium channel activity. However, the increase in a specific sodium cotransporter (NCC) observed in aldosterone-infused mice is likely an indirect effect of enhanced sodium channel activity and subsequent hypokalemia.

Summary: This article discusses the adaptive mechanisms involved in maintaining extracellular potassium concentration and membrane potential stability, including the signaling mechanisms in muscles and kidneys, the sensing and regulation of potassium concentration in skeletal muscles, the effects of potassium on aldosterone synthesis, and the kidney's response to changes in potassium concentration. The concept of sexual dimorphisms in renal potassium handling adaptation is introduced, and the molecular mechanisms underlying the cardiovascular benefits of a potassium-rich diet are discussed.

Aquaporins play a key role in water reabsorption.

High baseline ROMK activity in the mouse late distal convoluted and early connecting tubule probably contributes to aldosterone-independent K+ secretion

Viatcheslav Nesterov et al.

Summary: The renal outer medullary K+ channel (ROMK) and the epithelial Na+ channel (ENaC) are co-localized in the late distal convoluted tubule (DCT2), connecting tubule (CNT), and cortical collecting duct (CCD). Under conditions with low plasma aldosterone, ROMK predominantly functions in the DCT2/CNT. The high ROMK activity in the DCT2/CNT is critical for maintaining aldosterone-independent renal K+ secretion.

Summary: The study revealed that the activity of ENaC in the distal nephron of mouse kidney varies under different physiological conditions, while ROMK channels remain relatively stable. Conversely, increased potassium intake leads to recruitment of more secretion pathways to maintain potassium balance.

Critical role of the mineralocorticoid receptor in aldosterone-dependent and aldosterone-independent regulation of ENaC in the distal nephron

Viatcheslav Nesterov et al.

Summary: This study demonstrates that the activity of the epithelial Na+ channel (ENaC) in the aldosterone-sensitive distal nephron is a combination of aldosterone-dependent and aldosterone-independent mechanisms. The mineralocorticoid receptor (MR) is crucial for maintaining aldosterone-independent ENaC activity in the distal nephron, possibly through glucocorticoid activation and low expression of 11 beta-hydroxysteroid dehydrogenase 2 (11 beta-HSD2). The site-specific regulation of ENaC has significant physiological implications for renal sodium and potassium homeostasis.

ENaC expression correlates with the acute furosemide-induced K+ excretion

Niklas Ayasse et al.

Summary: This study found that the magnitude of acute furosemide-induced kaliuresis varies greatly depending on the basal molecular ENaC activity levels. K+ excretion was greatly enhanced in animals with increased ENaC expression under high K+ or low Na+ diet conditions, while it may be absent in animals fed a high Na+ diet. These results suggest that acute ENaC-dependent K+ excretion relies on both Na+ as substrate and molecular activation of ENaC.
