Summary: This study investigates the surprising reactivity patterns in Bronsted acid-mediated cyclizations of pyrrole substrates with pendant Michael acceptors. Experimental and theoretical studies show the significant influence of substituent effects on the viability of these transformations. Electronic effects and barrier-lowering interactions within transition states are identified as the reasons for the exclusive preference for 7-endo-trig cyclizations over 6-exo-trig cyclizations.

Summary: A diverse array of Stemona alkaloids were synthesized through tailored dyotropic rearrangements of beta-lactones, leading to various tricyclic and polycyclic structures with high efficiency, chemoselectivity, and stereospecificity. Additionally, a novel visible-light photoredox-catalyzed formal [3+2] cycloaddition was developed for accessing oxaspirobutenolide and related scaffolds.

Unified Total Syntheses of (±)-Sessilifoliamides B, C, and D

Summary: The total syntheses of Stemona alkaloids sessilifoliamides B and D, as well as the second synthesis of sessilifoliamide C, were successfully completed from a simple pyrrole substrate. The bicyclic lactam core was prepared on a gram scale via a Bronsted acid mediated cyclization and controlled oxidation with Dess-Martin periodinane. Yield percentages for the synthesis of sessilifoliamides B, C, and D were 24%, 13%, and 17% respectively, over multiple steps.

Revised Structures of Dehydrostenines A and B: Total Syntheses of (±)-Dehydrostenine A and Structure Assigned to Dehydrostenine B

Summary: The first total syntheses of the Stemona alkaloid dehydrostenine A and dehydrostenine B have been completed. Two independent Bronsted-acid-mediated intramolecular Michael additions were used to construct the tetracyclic dehydrostenine core. Revisions of the structures originally assigned to dehydrostenines A and B are proposed based on synthetic studies and literature analysis.

Synthesis of the Proposed Structures of Parvistemoamide and Their Transformations to Stemoamide Derivatives

Summary: The proposed structures of parvistemoamide have been achieved by macrolactamization, but characterization data of synthetic samples did not match with those of the natural sample. The transformation of the highly strained 10-membered lactam ring in parvistemoamide into the pyrrolo[1,2-alpha]-azepine nucleus in stemoamide is accomplished for the first time by either transannular cyclization or Pilli's transformation. This research may promote the total synthesis of other more complex stemoamide-type or medium-sized-ringcontaining Stemona alkaloids.
