Cryo-EM structures reveal high-resolution mechanism of a DNA polymerase sliding clamp loader

Christl Gaubitz et al.

Summary: In this study, the structures of Saccharomyces cerevisiae clamp loader Replication Factor C (RFC) bound to its sliding clamp Proliferating Cell Nuclear Antigen (PCNA) were described, revealing the mechanisms of opening and closing the sliding clamp and the exploration of primer-template junction using a base-flipping mechanism. ATP hydrolysis is triggered by interactions with both PCNA and DNA, explaining the switch-like ATPase activity of RFC. This work provides important insights into the DNA replication mechanism and cellular biology.

DNA is loaded through the 9-1-1 DNA checkpoint clamp in the opposite direction of the PCNA clamp

Fengwei Zheng et al.

Summary: The Cryo-EM analysis shows that Rad24-RFC loads the 9-1-1 clamp onto 5'-recessed DNA to activate the DNA damage checkpoint. This loading mechanism is different from RFC, and involves a unique way of threading DNA into the clamp.

Dari Kimanius et al.

Summary: This article introduces new tools for processing cryo-EM images in the fourth major release of the RELION software, including VDAM algorithm, convolutional neural network, and a framework for executing multiple jobs, along with a utility called MDCatch for metadata gathering during microscope data acquisition. These tools aim to provide fast and robust procedures for unsupervised cryo-EM structure determination, with potential applications for on-the-fly processing and flexible, high-throughput structure determination pipelines.

Phosphorylation-dependent assembly of DNA damage response systems and the central roles of TOPBP1

Matthew Day et al.

Summary: The cellular response to DNA damage involves extensive post-translational modifications of proteins, particularly through targeted phosphorylation. Understanding phosphorylation-mediated interactions helps define pathways coordinating DDR and identify new therapeutic targets. This review focuses on how phosphorylated motifs are recognized by BRCT domains in DDR proteins, with a specific interest in TOPBP1.

Structural basis for recruitment of the CHK1 DNA damage kinase by the CLASPIN scaffold protein

Matthew Day et al.

Summary: CHK1 is a protein kinase that phosphorylates multiple targets downstream of activated ATR, and its interaction with CLASPIN is essential for its activation by ATR. The crystal structure of CHK1 bound to a high-affinity motif from CLASPIN reveals the mechanism of how CLASPIN functions as a scaffold for CHK1 activation.
