A genetic screen in C. elegans reveals roles for KIN17 and PRCC in maintaining 5' splice site identity

Summary: Pre-mRNA splicing, carried out by spliceosomes, is a crucial step in eukaryotic gene expression. The mechanism by which splice site identity is maintained during dynamic changes in the spliceosome is not fully understood. Through a genetic screen in the nematode Caenorhabditis elegans, researchers identified KIN17 and PRCC as factors involved in splice site choice. Mutations in these factors promote the usage of an unusual proximal 5' splice site, changing the alternative splice site usage at native sites genome-wide. This study provides insights into the mechanisms by which the spliceosome maintains splice site identity.

SMG5-SMG7 maintain NMD activity by enabling SMG6 activation. The loss of SMG5-SMG7-dependent pathway can also inactivate the SMG6-dependent branch, indicating a functional connection between the final NMD steps.
