Modeling and targeting of erythroleukemia by hematopoietic genome editing

Ilaria Iacobucci et al.

Summary: Multiplexed genome editing in mouse hematopoietic cells led to the development of preclinical models for acute erythroid leukemia (AEL) and non-erythroid acute leukemia, highlighting the central role of mutational cooperativity in phenotype determination and clonal evolution. Clonal expansion during tumor evolution was driven by mutational cooccurrence, and mouse and human AEL exhibited deregulation of genes regulating erythroid development. Drug sensitivity was associated with the leukemia genotype, showing therapeutic potential for specific combinations of mutations in AEL.

Histone acetylation dynamics modulates chromatin conformation and allele-specific interactions at oncogenic loci

Stephanie Sungalee et al.

Summary: The study demonstrates that the abundance of H3K27ac affects the chromatin conformation of enhancer-promoter interacting regions, and depletion of H3K27ac alters the frequency and spreading of these interactions, thereby impacting oncogene expression. Enhancer hijacking mediated by chromosomal translocations influences the epigenetic status of specific regions, leading to distinct intrachromosomal interactions in homologous chromosomes.

CHIPIN: ChIP-seq inter-sample normalization based on signal invariance across transcriptionally constant genes

Lelia Polit et al.

Summary: CHIPIN is a new R package that allows for the normalization of ChIP-seq signals using gene expression data when spike-in information is not available. The method provides graphs and statistics to assess normalization efficiency and antibody specificity. Additionally, CHIPIN can be used for open chromatin ATAC-seq or DNase hypersensitivity data.

HDAC1 is required for GATA-1 transcription activity, global chromatin occupancy and hematopoiesis

Bowen Yan et al.

Summary: The activity of hematopoietic factor GATA-1 is modulated through acetylation and deacetylation processes mediated by p300/CBP and HDAC1/2 respectively. The deacetylation of GATA-1 is crucial for gene activation, chromatin binding, and the commitment and differentiation of erythroid/megakaryocyte cells. Mutations preventing GATA-1 deacetylation result in impaired erythroid differentiation and abnormalities in cell subpopulations and gene expression patterns in hematopoietic stem cells and progenitors.
