Experimental and modeling results show that these bundles are embedded in a continuous and contractile network, and the propagation of their contraction throughout the cell is dependent on this embedding. The mechanism of stress fibre assembly through the coalescence of actin filaments in the cell cortex enables the transmission of mechanical forces throughout the entire cell.

Generation of stress fibers through myosin-driven reorganization of the actin cortex

These findings demonstrate a new mechanism for de novo generation of stress fibers from the actin cortex, highlighting the role of stochastic myosin pulses in the assembly of functional actomyosin bundles.

Mechanism of actin filament nucleation

A barbed end interference mechanism reveals how capping protein promotes nucleation in branched actin networks

Summary: Heterodimeric capping protein (CP/CapZ) is crucial for the assembly of branched actin networks by both preventing filament elongation and promoting nucleation. Structural features of CP, particularly the C-terminal tentacle extension, play a key role in indirectly stimulating the activity of Arp2/3 activating nucleation promoting factors (NPFs) by preventing their association with filament barbed ends. Deletion of the beta tentacle modestly impairs capping, but strongly inhibits nucleation promoting factors' function in growing branched actin networks, revealing a mechanism by which CP balances capping and nucleation processes.
