Histone editing elucidates the functional roles of H3K27 methylation and acetylation in mammals

Summary: This study investigated the regulatory role of histone H3 modifications on gene expression using CRISPR base editing in mouse embryonic stem cells. The results showed that H3K27 is an essential substrate for PRC2, while other modifications besides H3K27 acetylation may be involved in mediating CBP/EP300 function.

Summary: Histone acetylation, a common feature of transcription, is largely dependent on transcription due to the recruitment and activity of histone acetyltransferases by RNA polymerase II. Promoter-bound HATs are unable to acetylate histones without active transcription, indicating that HAT activity is regulated post-recruitment.
