Discovery of spirooxindole-ferrocene hybrids as novel MDM2 inhibitors

Summary: A series of spirooxindole-ferrocene hybrids with five or four contiguous chiral centers were designed and synthesized through organocatalysis. Compound 5d was identified as the most potent MDM2 inhibitor, suppressing MDM2-mediated p53 degradation, inducing apoptosis, and promoting oxidative damage. Molecular docking studies have shown that 5d binds to MDM2 by mimicking the Trp23 and Leu26 residues of p53, providing a basis for the development of novel multifunctional MDM2 inhibitors.

Summary: The study successfully synthesized bi-spirooxindole-engrafted rhodanine analogs with excellent stereo-selectivity and high yield through a one-pot multicomponent [3 + 2] cycloaddition reaction. X-ray crystallographic investigations and Hirshfeld calculations revealed the crystal structure and intermolecular interactions, while theoretical study was conducted on the mechanism of the 32CA reaction.

Synthesis, X-ray structural determination and biological evaluation of novel ferrocene grafted spiroquinoxalinopyrrolidine

Summary: A hybrid heterocycle comprising ferrocene, spiropyrrolidine, and quinoxaline structural moieties was synthesized in good yield via ionic liquid expedited multicomponent 1,3-dipolar cycloaddition reaction. Structural properties were studied using single crystal X-ray crystallographic studies, while Fe-C interactions and antimicrobial potential were investigated through NBO and AIM calculations. The compound showed significant antimicrobial activity against E. coli and S. enterica, as well as antioxidant activity with an IC50 value of 33.80 μg/mL.

Design, Synthesis, Chemical and Biochemical Insights Into Novel Hybrid Spirooxindole-Based p53-MDM2 Inhibitors With Potential Bcl2 Signaling Attenuation

Summary: Novel hybrid spirooxindole-based p53-MDM2 inhibitors with Bcl2 signaling attenuation were synthesized and shown to exhibit anticancer effects through apoptosis induction and modulation of p53 and Bcl2 gene expressions. The synthesized compounds demonstrated superior cytotoxic activities compared to traditional chemotherapeutic drugs, suggesting their potential as effective anticancer agents. Docking simulations provided insights into the possible binding modes of the compounds within MDM2.

Compounds 8i and 8y showed the strongest acetylcholine esterase inhibition and were further studied with molecular docking for their interaction with the active site of hAChE.
