The rise and fall of anandamide: processes that control synthesis, degradation, and storage

Summary: Anandamide, an endocannabinoid derived from arachidonic acid-containing membrane lipids, exerts its effects primarily through CB1, CB2, and TRPV1 receptors, participating in the regulation of sleep, eating, pleasure enhancement, and pain relief. This manuscript reviews anandamide synthesis, degradation, and storage, and discusses the homeostasis of the anandamide signaling system.

The effect of age and sex on the expression of GABA signaling components in the human hippocampus and entorhinal cortex

Summary: The study found age- and sex-related alterations in GABA(A/B)R subunits in the ECx and hippocampus, suggesting that these differences may significantly impact GABAergic neurotransmission and disease susceptibility or progression. The expression levels of GAD 65/67, however, were not significantly affected by age or sex. These results provide important insights into the regulation of GABA signaling components in the brain.

Additionally, a high content of monounsaturated fatty acids in edible oils may improve lipid profiles and reduce damage caused by oxidized lipoproteins and free radicals.

Noncanonical Activity of Endocannabinoids and Their Receptors in Central and Peripheral Synapses

Summary: This review discusses new aspects of endocannabinoid functions including biased agonism of endogenous and synthetic cannabinoids, coupling of CB receptors with various G-proteins and beta-arrestins, as well as noncanonical tonic activity of endocannabinoids. The role of endocannabinoids in neurotransmitter release, short-term and long-term potentiation, and synaptic function is highlighted, with the conclusion that the endocannabinoid system has a wider range of modulating effects than previously recognized. Re-evaluation of its functions considering noncanonical features will lead to a better understanding of its role in normal and pathological nervous system functioning.

Epilepsy with myoclonic-atonic seizures (Doose syndrome): Clarification of diagnosis and treatment options through a large retrospective multicenter cohort

Summary: This study presented the largest cohort of patients with EMAS to date, revealing that global developmental delay was present in nearly half of the children during the course of the disease. Most children experienced seizures after the age of 2, with EEG showing abnormal findings in the majority. Diet therapy was found to be significantly more effective than the first-line antiseizure drugs in achieving seizure reduction.
