Summary: Skin, the largest organ in the human body, plays a crucial role in protecting against external agents. Maintaining a healthy skin micro-environment relies on the interaction between skin-resident microbiota, structural cells, and immune cells. Environmental stressors and genetic factors can disrupt this delicate balance, leading to inflammatory conditions. Recent studies have highlighted the importance of immune cells, specifically Type-2 immunity, in skin resolution, repair, and immune dysregulation. Interleukin receptor alpha (IL-4R alpha) is a key component of Type-2 immunity and a target for inflammatory skin conditions. This review discusses IL-4R alpha immunity, genetics, and signaling in pre-clinical models of atopic dermatitis.
