Formate dehydrogenases for CO 2 utilization

Summary: New measures are urgently needed to reduce atmospheric CO2 levels. Formate dehydrogenases (FDHs) have the potential to catalyze the conversion of CO2 to formate, providing a novel first step approach for biocatalytic carbon capture and utilization. However, improvements are needed in the efficacy of the electron donor, enzyme stabilization, and enhancing the desired reverse FDH reactivity. Recent research has shown that the commonly used cofactor NADH is inefficient for FDH catalyzed CO2 conversion. Understanding the details of the redox reactions and structure-function relations of both metal dependent and metal-independent FDHs is crucial for promoting enzymatic CO2 utilization.

Development of an Ionic Porphyrin-Based Platform as a Biomimetic Light-Harvesting Agent for High-Performance Photoenzymatic Synthesis of Methanol from CO2

Summary: NADH is essential for enzymatic hydrogenation of CO2 to methanol, but its high costs limit its large-scale application. Photocatalytic reduction of NAD(+) to NADH is a promising solution utilizing solar energy. Efficient electron transfer from the photosensitizer to the catalyst is crucial for the success of NADH regeneration. Ionic porphyrin was identified as the most efficient photosensitizer for in situ NADH regeneration, leading to a sevenfold increase in methanol concentration when integrated with enzyme cascade reactions using a membrane support.

Improved biocatalytic cascade conversion of CO2 to methanol by enzymes Co-immobilized in tailored siliceous mesostructured cellular foams

Summary: In this study, CO2 was enzymatically reduced to methanol in a cascade reaction involving three enzymes: formate-, formaldehyde- and alcohol dehydrogenase. By co-immobilizing the three dehydrogenases in siliceous mesostructured cellular foams (MCF), the yield of this reaction was significantly improved. Functionalization of the silica material with mercaptopropyl groups enhanced the interaction between the enzymes and support, leading to a 4.5-fold increase in methanol yield compared to enzymes free in solution.
