Amyotrophic Lateral Sclerosis-Frontotemporal Dementia Shared and Divergent Neural Correlates Across the Clinical Spectrum

Summary: This study investigates the neural correlates in patients with amyotrophic lateral sclerosis (ALS) and behavioral variant of frontotemporal dementia (bvFTD) using MRI connectomic data. The results suggest that patients with ALS-ci/bi exhibit similar structural alterations as ALS-cn, but with enhanced functional connectivity in sensorimotor areas and decreased functional connectivity in the bvFTD-like pattern. These findings support the hypothesis that ALS-ci/bi may be considered as a phenotypic variant of ALS.

Structural MRI Signatures in Genetic Presentations of the Frontotemporal Dementia/Motor Neuron Disease Spectrum

Summary: Cortical, subcortical, and cerebellar gray matter atrophy was assessed in patients with frontotemporal lobar degeneration (FTLD) spectrum disorders using MRI. The study found that genetic FTD patients showed more severe and diffuse GM atrophy, with C9orf72 and GRN mutation carriers exhibiting the most extensive cortical volume loss. Different genetic mutations in FTLD patients showed variations in GM volume loss, which may serve as useful markers for gFTLD, particularly C9orf72-related disorders.

Impaired recognition of disgust in amyotrophic lateral sclerosis is related to basal ganglia involvement

Summary: In this study, emotion recognition in pure motor ALS patients was investigated. It was found that ALS patients had impaired disgust recognition, which was related to reduced volume of the left pallidum and lower cognitive performance. This suggests that impaired disgust recognition may be an early marker of cognitive decline in ALS patients without cognitive or behavioral impairment.
