Chromosome-specific telomere lengths and the minimal functional telomere revealed by nanopore sequencing

Samantha L. Sholes et al.

Summary: We developed a method to tag and measure telomere length in yeast using nanopore sequencing. We observed stable chromosome-specific telomere lengths and examined the role of RIF1 and TEL1 in telomere length regulation. The study suggests the influence of Y ' elements on telomere length regulation and provides insights into possible new biological mechanisms.

John Mascarenhas et al.

Summary: Imetelstat, a first-in-class telomerase inhibitor, has shown promising clinical benefits in higher risk myelofibrosis (MF) patients. This article outlines the rationale and design for the ongoing phase III trial, IMpactMF, which aims to compare the efficacy of imetelstat with the best available therapy (BAT) in MF patients refractory to JAK inhibitors. The primary objective is to assess overall survival, while secondary objectives include symptom and spleen responses, progression-free survival, and safety. Biomarker, cytogenetics and mutation analyses will also be performed.

Rap1 regulates TIP60 function during fate transition between two-cell-like and pluripotent states

Raymond Mario Barry et al.

Summary: This study uncovers the mechanism of gene expression regulation by the telomere binding protein Rap1. Rap1 interacts with the TIP60/p400 complex and modulates its histone acetyltransferase activity. Deletion of Rap1 increases the fraction of two-cell-like cells in mouse embryonic stem cells and leads to upregulation of certain genes.

Summary: This study reveals the structure of human telomerase and its interaction with TPP1. The binding of TPP1 to TERT activates telomerase activity. Additionally, the study found that elements in TERT and TER involved in template and telomeric DNA handling are structurally similar to those in Tetrahymena telomerase, providing unique insights into the mechanism of telomerase activity.

A novel p53 regulator, C16ORF72/TAPR1, buffers against telomerase inhibition

Yahya Benslimane et al.

Summary: A new gene C16ORF72/TAPR1 has been discovered to regulate telomere integrity and p53 levels, with cells lacking TAPR1 or TERT showing elevated p53 levels and sensitivity to nutlin-3a treatment, which can be alleviated by loss of p53.

Homologous recombination-mediated irreversible genome damage underlies telomere-induced senescence

Sabrina Ghadaouia et al.

Summary: Loss of telomeric DNA leads to telomere uncapping and triggers a persistent, p53-centric DNA damage response that sustains a stable senescence-associated proliferation arrest. Dysfunction of telomeres during cell division results in sporadic telomeric sister chromatid fusions and genome instability, ultimately leading to senescence mediated by p53. Blocking Rad51/RPA-mediated homologous recombination can prevent senescence despite dysfunctional telomeres, suggesting that repairing telomeres in the pre-senescent state could potentially prevent senescence and genome instability.

Diogo Paramos-de-Carvalho et al.

Summary: Cellular senescence is a complex and programmed cellular state with diverse physiological and pathological roles across the lifespan of an organism. New tools have allowed researchers to uncover the functional impact of senescent cells in various contexts, although reconciling these effects can be challenging. Ongoing clinical research is exploring the targeting of senescent cells and its potential implications in this rapidly evolving field.

Life Expectancy in Marine Mammals Is Unrelated to Telomere Length but Is Associated With Body Size

Kittisak Buddhachat et al.

Summary: This study used skin tissue samples from 23 marine mammal species to investigate the relationship between adult body weight, length, relative telomere length, and lifespan. The results showed correlations between lifespan and adult body weight and length, but no correlation with relative telomere length. Larger marine mammals were found to live longer, but relative telomere length was not a good predictor of species longevity.

Heritable variation in telomere length predicts mortality in Soay sheep

Summary: The study found that in wild Soay sheep, there was no significant association between telomere length and subsequent survival, suggesting that telomere length is not an important marker of biological aging or exposure to environmental stress in this study system. Instead, individual differences in average telomere length were associated with increased lifespan, indicating a genetic basis for this correlation.
