Physiological and pathological roles of protein kinase A in the heart

Yuening Liu et al.

Summary: Protein kinase A (PKA) serves as a central regulator of cardiac performance and morphology. Abnormal activation or inactivation of PKA is associated with a wide range of heart diseases, and PKA could be a promising drug target for cardioprotection. Therefore, specific PKA inhibitors or activators may represent valuable drug candidates for the treatment of heart diseases.

Pros and Cons of Pharmacological Manipulation of cGMP-PDEs in the Prevention and Treatment of Breast Cancer

Patrizia Di Iorio et al.

Summary: The cyclic nucleotides, cAMP and cGMP, act as second messengers in both normal and tumor cells, playing a crucial role in translating extracellular signals to intracellular responses. Inhibition of cGMP-hydrolyzing phosphodiesterases (PDEs) has shown potential as a therapeutic approach for cancer by regulating cell growth, apoptosis, and sensitivity to chemotherapy. However, the use of PDE5 inhibitors in breast cancer treatment remains controversial.

Summary: This study investigates the major features of PDE9A and PDE10A using computational methods, revealing the crucial residues for inhibitor binding affinity and selectivity. It is found that PDE10A selective inhibitors require interactions with more conservative residues compared to PDE9A. This study provides valuable insights for the design of selective inhibitors targeting PDE9A and PDE10A.

Inhibition of Colon Cancer Cell Growth by Phosphodiesterase Inhibitors Is Independent of cGMP Signaling

Yali Hou et al.

Summary: The use of phosphodiesterase (PDE) inhibitors for colorectal cancer (CRC) prevention and treatment has been of interest. This study finds that PDE inhibitors do not inhibit growth and viability of CRC cell lines by increasing cGMP and activating PKG as previously thought. The lack of endogenous guanylyl cyclase activity in colon cancer cells prevents PDE inhibitors from activating cGMP signaling. The inhibitory effect of PDE inhibitors on CRC growth is due to toxic effects at clinically unobtainable drug concentrations.

Genetic Alterations in Benign Adrenal Tumors

Georgia Pitsava et al.

Summary: The genetic basis of most types of adrenal adenomas has been studied, revealing associations with specific molecular defects. Genetic variants affecting the protein kinase A (PKA) signaling pathway have been linked to adrenal gland pathologies. Additionally, genes related to aldosterone production have been identified as causes of primary aldosteronism.

Long Term Survivals in Aggressive Primary Brain Malignancies Treated With an Adjuvant Ketogenic Diet

Kenneth A. Schwartz et al.

Summary: Aggressive primary brain tumors such as glioblastoma multiforme and grade IV astrocytoma are typically treated with surgery, radiation therapy, and chemotherapy. The addition of a ketogenic diet to rodents with brain tumors has shown promising results in increasing survival. In this study, adult patients with APBT underwent neurosurgery followed by an adjuvant ketogenic diet concurrent with radiation therapy and temozolomide. Younger patients showed longer survival rates compared to older patients. Further research is needed to confirm these findings, particularly in younger patients under the age of 40.
