Urolithins: a Comprehensive Update on their Metabolism, Bioactivity, and Associated Gut Microbiota

Rocio Garcia-Villalba et al.

Summary: Urolithins, metabolites produced by the gut microbiota from polyphenols, have been extensively studied for their potential health benefits. Recent research has identified 13 urolithins and their corresponding conjugated metabolites in various human fluids and tissues. The study highlights the importance of understanding the microorganisms involved in urolithin production and the composition of gut microbiota associated with urolithin metabolism. However, further in vitro studies are needed to confirm the reported activities of urolithins.

Urolithin A Attenuates Diabetes-Associated Cognitive Impairment by Ameliorating Intestinal Barrier Dysfunction via N-glycan Biosynthesis Pathway

Yao Xiao et al.

Summary: This study investigates the effect of Urolithin A (UA) on diabetes-associated cognitive impairment in a mouse model of type 2 diabetes mellitus (T2DM) induced by a high-fat diet (HFD) and streptozotocin (STZ). The results show that UA treatment can attenuate cognitive impairment and reduce levels of metabolic endotoxemia and proinflammatory cytokines in serum. UA also suppresses gut/brain inflammation, improves gut barrier function, and restores the dysregulation of genes involved in N-glycan biosynthesis. Unlike metformin, the beneficial effects of UA are independent of the gut microbiome and short-chain fatty acids. These findings suggest that UA has the potential to be a novel pharmacological target for diabetes therapy by manipulating the gut-brain axis and N-glycan metabolism.

Human gut bacteria produce TH17-modulating bile acid metabolites

Donggi Paik et al.

Summary: This study identifies bile acids produced by gut bacteria that inhibit the differentiation of T(H)17 cells, a key immune cell type involved in inflammatory disorders such as inflammatory bowel disease. The levels of these bile acids are reduced in patients with inflammatory bowel disease and are inversely correlated with the expression of T(H)17-cell-associated genes.

Urolithin A protects against acetaminophen-induced liver injury in mice via sustained activation of Nrf2

Zhimin Gao et al.

Summary: Urolithin A (UA), a metabolite of ellagitannin natural products, protects against acetaminophen-induced liver injury (AILI) and is superior to N-acetylcysteine (NAC) in dosage and therapeutical time window. UA promotes mitophagy and activated Nrf2/ARE signaling in the liver, leading to alleviation of acetaminophen-induced oxidative stress and hepatic necrosis.

Polyphenol-Mediated Gut Microbiota Modulation: Toward Prebiotics and Further

Maria Carolina Rodriguez-Daza et al.

Summary: The genome of gut microbes encodes enzymes that metabolize unabsorbed (poly)phenols, impacting host health. While certain probiotics transform these compounds into bioactive metabolites, other gut symbionts benefit from (poly)phenols without metabolizing them. This dual microbial effect, termed "duplibiotic," promotes beneficial gut bacteria and may have therapeutic potential for metabolic disturbances and gut dysbiosis.
