Summary: This study demonstrates that Clostridioides difficile spores can adhere to and be phagocytosed by macrophages, with only spores from toxigenic strains significantly stimulating inflammatory cytokine production and inducing cytotoxicity. Spores from isogenic TcdA and TcdB double mutant strains induce lower inflammatory responses and cytotoxicity in macrophages, which can be restored by pre-exposure to toxins. These findings suggest a potential role of C. difficile toxins coating spores during sporulation, impacting the pathogenesis of C. difficile in vivo.
