A Two-Pore Physiologically Based Pharmacokinetic Model to Predict Subcutaneously Administered Different-Size Antibody/Antibody Fragments

Two-pore physiologically based pharmacokinetic model validation using whole-body biodistribution of trastuzumab and different-size fragments in mice

Summary: The two-pore PBPK model proposed by the lab for protein therapeutics of different sizes was validated using plasma PK data and tissue distribution data of different-size antibodies. The model showed relatively accurate predictions for plasma, tissues, and tumor exposure of studied molecules, proving its potential as a platform for developing a generic PBPK model for protein therapeutics in the future.

Summary: Advances in antibody engineering have allowed the construction of novel molecular formats, providing new opportunities for cancer immunotherapy drug discovery, but also revealing limitations in knowledge of structure-activity relationships. By constructing a representative library and using different shapes and hinge flexibilities of IgG-like formats, it was found that one-armed antibodies have higher renal uptake compared to bivalent antibodies.
