Chloroquine combined with concurrent radiotherapy and temozolomide for newly diagnosed glioblastoma: a phase IB trial

Inge Compter et al.

Summary: Treatment with chloroquine in combination with radiotherapy and concurrent temozolomide for newly diagnosed glioblastoma showed favorable toxicity and promising overall survival. The maximum tolerated dose was determined to be 200 mg of chloroquine daily. Further clinical studies are supported by these results.

Combined Treatment with Acalabrutinib and Rapamycin Inhibits Glioma Stem Cells and Promotes Vascular Normalization by Downregulating BTK/mTOR/VEGF Signaling

Yu-Kai Su et al.

Summary: The study evaluated the effects of BTK and mTOR inhibition on glioblastoma (GBM) and found a synergistic relationship between acalabrutinib and rapamycin, which effectively suppressed CSC phenotypes and angiogenesis potential, promoting vascular normalization for chemoresistant/radioresistant GBM.

MSC-AS1 knockdown inhibits cell growth and temozolomide resistance by regulating miR-373-3p/CPEB4 axis in glioma through PI3K/Akt pathway

Chong Li et al.

Summary: In TMZ-resistant glioma, high expression of MSC-AS1 was found to be associated with reduced overall survival rate, while inhibition of MSC-AS1 decreased IC50 value of TMZ, promoted apoptosis, and affected PI3K/Akt pathway. Further investigation revealed that MSC-AS1 acted as a miR-373-3p sponge and miR-373-3p targeted CPEB4 directly, influencing TMZ sensitivity. Silencing miR-373-3p reversed the promoting effect of MSC-AS1 or CPEB4 knockdown on TMZ sensitivity, indicating a potential regulatory mechanism for chemoresistance.

Current FDA-Approved Therapies for High-Grade Malignant Gliomas

Jacob P. Fisher et al.

Summary: The standard of care for high-grade gliomas includes surgical resection, radiation therapy, and temozolomide, which have improved patient survival rates. Besides temozolomide, there are several other FDA-approved treatments for HGG, with tumor treatment fields being the only one shown to significantly improve survival rates.

Molecular Dosimetry of Temozolomide: Quantification of Critical Lesions, Correlation to Cell Death Responses, and Threshold Doses

Bjoern Stratenwerth et al.

Summary: Temozolomide (TMZ) is a DNA-methylating agent commonly used in cancer chemotherapy, particularly for glioblastoma multiforme (GBM). The study found that O(6)MeG, DNA double-strand breaks (DSB), and apoptosis all increase linearly with dose, while cellular senescence also shows a linear relationship without a threshold at therapeutically relevant dose levels. The genotoxic and cytotoxic effects observed suggest that the TMZ target concentration can trigger significant cytotoxic and cytostatic effects in vivo.

Rodent Models to Analyze the Glioma Microenvironment

Susann Hetze et al.

Summary: Animal models are crucial for studying glioma development and invasion, with preclinical approaches helping to assess treatment efficacy of new drugs. The tumor microenvironment has been recognized as a key player in the nature of glioma, and available rodent models are evaluated for their usability in researching the glioma microenvironment.

Effect of naringenin and its combination with cisplatin in cell death, proliferation and invasion of cervical cancer spheroids

Oswaldo Pablo Martinez-Rodriguez et al.

Summary: The flavonoid naringenin has shown cytotoxic effects on cervical cancer cells and may be considered as a coadjuvant in treatment. Naringenin impairs cell viability and induces cytotoxicity through necrotic cell death. Additionally, it enhances the anti-tumor effect of low concentrations of cisplatin by decreasing cell viability and invasion in HeLa spheroids, indicating its potential use as an adjuvant therapy against cervical cancer.
