Optimizing the diagnostic workflow for acute lymphoblastic leukemia by optical genome mapping

Katrina Rack et al.

Summary: Acute lymphoblastic leukemia (ALL) is a malignant disease that can be divided into distinct subtypes based on clinical and genomic features. Traditional diagnostic methods are laborious and costly. This study applied optical genome mapping (OGM) to diagnose ALL and found that OGM can accurately detect common genomic abnormalities, improve the detection rate and cytogenetic resolution, and reduce turnaround time.

Myelodysplastic/myeloproliferative neoplasms-unclassifiable with isolated isochromosome 17q represents a distinct clinico-biologic subset: a multi-institutional collaborative study from the Bone Marrow Pathology Group

Rashmi Kanagal-Shamanna et al.

Summary: The study revealed that patients with MDS/MPN-i(17q) were typically younger with lower platelet and neutrophil counts, commonly displayed bilobed neutrophils and had a higher mutation frequency. These patients had a poorer prognosis and shorter overall survival compared to those without i(17q).

Hybrid sequencing resolves two germline ultra-complex chromosomal rearrangements consisting of 137 breakpoint junctions in a single carrier

Jesper Eisfeldt et al.

Summary: This study reported a healthy female carrying two novel CCRs and characterized these complex rearrangements. The results were validated using multiple cytogenetic methods, revealing the highest number of breakpoint junctions (137) and indicating an association between CCR formation and active gene transcription.

Analytical and clinical performance of chromosomal microarrays compared with FISH panel and conventional karyotyping in patients with chronic lymphocytic leukemia

Zhenya Tang et al.

Summary: In this study, comparative analysis between CMA and traditional cytogenetic methods such as karyotyping and FISH revealed that CMA can provide additional analytical information but its clinical utility is limited to a small number of patients. Evaluation based on NCCN and CGC guidelines showed that combining FISH with CMA had higher capture rates of prognostic relevant cytogenetic information. Further research is needed to explore the potential of CMA as an alternative to karyotyping.

The Clinical Utility of Optical Genome Mapping for the Assessment of Genomic Aberrations in Acute Lymphoblastic Leukemia

Jonathan Lukas Luehmann et al.

Summary: This study investigates the feasibility of using Optical Genome Mapping (OGM) as an all-in-one approach to determine the genetic risk profile of childhood Acute Lymphoblastic Leukemia (ALL). OGM has the potential to simplify the diagnostic workflow and identify new structural variants helpful for classifying patients into treatment groups. The results show that OGM can detect both established and new potential prognostic markers in ALL, demonstrating its promise for future diagnostic applications.
