Cancer statistics, 2022

Rebecca L. Siegel et al.

Summary: Each year, the American Cancer Society compiles data on cancer occurrence and outcomes in the United States. The latest data shows that breast and prostate cancer progress has stagnated, while lung cancer has shown improvements in survival rates. Lung cancer incidence for advanced disease has declined while localized-stage rates have increased, resulting in higher survival rates. Mortality patterns align with incidence trends, with lung cancer deaths declining rapidly, breast cancer deaths slowing, and prostate cancer deaths stabilizing.

Multi-omics integration in biomedical research - A metabolomics-centric review

Maria A. Woerheide et al.

Summary: Recent advances in high-throughput technologies have enabled the profiling of multiple layers of a biological system, generating vast amounts of data that can be integrated in multi-omics studies. Integrative analysis of such datasets is complicated by the high dimensionality, heterogeneity, and lack of universal protocols. While previous reviews focused on integrating two omics layers with a phenotype, this review highlights methods to combine metabolomics data with two or more omics layers without focusing on a specific phenotype.

Multi-omics approaches in cancer research with applications in tumor subtyping, prognosis, and diagnosis

Otilia Menyhart et al.

Summary: Multi-omics data integration offers unparalleled opportunities for understanding complex diseases like cancer, but faces challenges such as uneven maturity of different omics approaches and limited data processing capacity. Efforts to standardize sample processing, train experts for data analysis, and close the gap between data generation and processing are crucial for the translatability of theoretical findings.

A Detailed Catalogue of Multi-Omics Methodologies for Identification of Putative Biomarkers and Causal Molecular Networks in Translational Cancer Research

Efstathios Iason Vlachavas et al.

Summary: Recent advancements in sequencing and biotechnological methodologies have resulted in the generation of large volumes of molecular data, which can be integrated with clinical information to discover links between biological processes and disease. Utilizing data-driven approaches for multi-omics data integration can lead to the identification of molecular networks impacting cancer pathophysiology, improving disease diagnosis, prognosis, prevention, and therapy. Computational methodologies and tools, such as Multi-Omics Factor Analysis (MOFA) and netDX, are tested in this study to gain biological knowledge from omics data and understand perturbations in distinct cancer types. The performed analyses yield meaningful and novel findings for translational cancer research and personalized therapy.

Hidden Targets in RAF Signalling Pathways to Block Oncogenic RAS Signalling

Aoife A. Nolan et al.

Summary: Oncogenic RAS mutations are key drivers of human cancers, making RAS inhibition a crucial goal in oncology. Efforts to block effector pathways like RAF-MEK-ERK have been largely disappointing, leading researchers to investigate RAF-dependent pathways independent of catalytic activity as potential targets for blocking oncogenic RAS signaling. The roles of RAF kinase-independent functions in apoptosis, cell cycle progression, and cell migration have been well documented.

Analysis of LAGEs Family Gene Signature and Prognostic Relevance in Breast Cancer

Hoang Dang Khoa Ta et al.

Summary: The LAGE family's expression in breast cancer is closely related to patient prognosis, with high expression of LAGE3 associated with poorer survival. LAGE family genes show significant associations with the cell cycle, focal adhesion, and extracellular matrix receptor interactions.
