Chemical Biology of Reactive Sulfur Species: Hydrolysis-Driven Equilibrium of Polysulfides as a Determinant of Physiological Functions

Tomohiro Sawa et al.

Summary: Polysulfide species, highly nucleophilic in nature, play a crucial role in antioxidant activities. Recent studies have shown that these species undergo hydrolysis to form nucleophilic hydropersulfide/polysulfide and electrophilic sulfenic acid species. Understanding the metabolism and occurrence of polysulfide species is critical for further exploration in this field.

Efficient inhibition of glyceraldehyde-3-phosphate dehydrogenase (GAPDH) by sulfuration with solubilized elemental sulfur

Sarah G. Bolton et al.

Summary: Hydrogen sulfide, carbon monoxide, and nitric oxide have attracted increasing scientific interest as gasotransmitter signaling molecules. Particularly, hydrogen sulfide has been studied for its ability to modulate cellular redox environments through the production of oxidized sulfur species. This study explores the inhibition of glyceraldehyde-3-phosphate dehydrogenase by pure sulfur and different polysulfides, demonstrating the potential of this simplified system for studying sulfur redox chemistry.

Stabilities of Three Key Biological Trisulfides with Implications for Their Roles in the Release of Hydrogen Sulfide and Bioaccumulation of Sulfane Sulfur

Eric M. Brown et al.

Summary: Trisulfides and higher polysulfides serve as important reservoirs of sulfane sulfur in the body and rapidly release persulfides, which have powerful antioxidant and gasotransmitter properties. However, their stabilities in aqueous systems and under different conditions remain poorly understood. This study synthesized biologically important trisulfides and a representative polysulfide and investigated their reactivity in buffered D2O. Results showed that tri- and tetrasulfides degrade rapidly in the presence of amines and are unstable under basic conditions. These findings have implications for understanding the role and stability of polysulfides in biological systems.

A Review of Chemical Tools for Studying Small Molecule Persulfides: Detection and Delivery

Kearsley M. Dillon et al.

Summary: Persulfides have emerged as reactive sulfur species with enhanced antioxidant abilities compared to hydrogen sulfide, but their instability and cross-reactivity hinder the elucidation of their precise biological roles. To facilitate the study of persulfides, chemists have designed various chemical tools and techniques, including persulfide trapping reagents and sensors, as well as persulfide donors that release persulfides in response to triggers.
