Evidence for a Long-Lived, Cu-Coupled and Oxygen-Inert Disulfide Radical Anion in the Assembly of Metallothionein-3 Cu(I)4-Thiolate Cluster

Jenifer S. Calvo et al.

Summary: Researchers have identified and characterized transient radical intermediates formed in the reaction between Zn7MT-3 and Cu(II). These intermediates are stable and have long lifetimes, providing insights into the assembly process of the MT-3 Cu(I)(4)-Cys(5) cluster.

Evidence for a Long-Lived, Cu-Coupled and Oxygen-Inert Disulfide Radical Anion in the Assembly of Metallothionein-3 Cu(I)(4)-Thiolate Cluster

Jenifer S. Calvo et al.

Summary: MT-3 protein reduces Cu(II) to Cu(I) and forms a multi-nuclear Cu(I)(4)-Cys(5-6) cluster, generating long-lived and oxygen-stable disulfide radical anion intermediates.

Intermediate Cu(II)-Thiolate Species in the Reduction of Cu(II)GHK by Glutathione: A Handy Chelate for Biological Cu(II) Reduction

Iwona Ufnalska et al.

Summary: GHK is a tripeptide with biological functions attributed to its copper-complexed form Cu(II)GHK, but little is known about its molecular mechanism of action. This study investigates the reaction of Cu(II)GHK with the strongest reductant in human plasma, GSH, using spectroscopic techniques and cyclic voltammetry to reveal important reaction intermediates. The findings suggest that the GSH-Cu(II)GHK complex may play a crucial role in copper trafficking as a precursor of Cu(I) ions.

Metal- and metalloid-based compounds to target and reverse cancer multidrug resistance

Andreia Valente et al.

Summary: Drug resistance remains a major cause of cancer treatment failure, especially in the late stage of the disease. Metal and metalloid compounds offer the possibility to design fine-tuned drugs to overcome drug-resistant cancer cells. This review highlights the importance of platinum drugs in clinics and discusses strategies to reverse resistance to platinum drugs and multidrug resistance based on ABC efflux pumps.

Modulation of Intracellular Copper Levels as the Mechanism of Action of Anticancer Copper Complexes: Clinical Relevance

Maria Babak et al.

Summary: Copper plays a crucial role in cellular growth and development and its imbalance can lead to toxicity. This review explores the mechanisms of intracellular copper balance interference in tumor development and progression, and discusses the potential of using copper-depleting and copper-overloading conditions for improved patient outcomes. Additionally, the advantages of pre-formed copper complexes in comparison to in situ formed complexes are highlighted.
