Engineered virus-like particles for efficient in vivo delivery of therapeutic proteins

Summary: We have developed and applied engineered DNA-free virus-like particles (eVLPs) for efficient delivery of gene editing tools, achieving high levels of base editing. By overcoming the limitations of cargo packaging, release, and localization, the fourth-generation eVLPs demonstrated efficient base editing in mouse and human cells. In addition, the off-target effects of eVLPs were significantly reduced compared to other delivery methods such as AAV or plasmid.

Summary: This study elucidates the crucial role of HIV-1 Gag in viral assembly and the structural features of partial CA hexamers at lattice edges. Molecular dynamics simulations and free energy calculations reveal that partial hexamers are more prone to helix-to-coil transitions compared to complete six-helix bundles.

A multiscale coarse-grained model of the SARS-CoV-2 virion

Summary: This article presents the current status and ongoing development of a largely bottom-up coarse-grained (CG) model of the SARS-CoV-2 virion. The CG model was constructed using data from cryo-electron microscopy, x-ray crystallography, and computational predictions to build molecular models of structural SARS-CoV-2 proteins, which were then assembled into the complete virion model. The CG model allows for capturing viral behavior that is difficult to observe in atomistic simulations and can be iteratively improved with new data.

Structure of the mature Rous sarcoma virus lattice reveals a role for IP6 in the formation of the capsid hexamer

Summary: IP6 serves as a more potent assembly cofactor for RSV than for HIV-1, and its removal in cells significantly reduces RSV infectivity. Visualized structures of mature capsids show IP6-like density in the CA hexamer, coordinated by lysines and arginines, with phosphate and IP6 affecting CA assembly differently. The flexibility of CA hexamers and rigidity of pentamers determine the variable shape of CA polyhedrons in mature RSV particles.
