The capability of coenzyme Q10 to enhance heat tolerance in male rabbits: evidence from improved semen quality factor (SQF), testicular oxidative defense, and expression of testicular melatonin receptor MT1

A. I. El-Sayed et al.

Summary: This study aimed to investigate the antioxidative effect of CoQ10 on sperm quality, testicular antioxidant activities, and male fertility under heat stress. The results showed that CoQ10 supplementation had significant effects on semen quality factor, testicular antioxidant activities, concentration of testosterone, integrity of testicular DNA, and the expression of melatonin receptors, ultimately leading to an improvement in buck fertility and increased prolificacy in females when inseminated with treated bucks. CoQ10 tends to decrease oxidative stress and enhance testicular antioxidant activities, making it a suitable feed supplement to increase fertility and reduce the harmful effects of heat stress in male rabbits.

Regulation of cancer cell glucose metabolism is determinant for cancer cell fate after melatonin administration

Carmen Rodriguez et al.

Summary: Multiple oncogenic pathways and local microenvironmental conditions converge on regulating cancer cell metabolism, with melatonin considered a therapeutic target due to its reported antitumor effects. High concentrations of melatonin have shown effectiveness in reducing cell growth and inducing cell death in certain types of tumors, suggesting its potential role in regulating glucose metabolism in cancer cells.

Energy substrate metabolism and mitochondrial oxidative stress in cardiac ischemia/reperfusion injury

Maija Dambrova et al.

Summary: The heart is highly flexible in its metabolism and mitochondria play a crucial role in oxidative phosphorylation and heart function. ROS production pathways during ischemia/reperfusion involve glucose, fatty acids, and the Krebs cycle, with strategies available to modulate substrate accumulation for ROS production mitigation.

Increased demand for NAD+ relative to ATP drives aerobic glycolysis

Alba Luengo et al.

Summary: Increasing the activity of the pyruvate dehydrogenase complex impairs cell proliferation by reducing the NAD(+)/NADH ratio, which is caused by increased mitochondrial membrane potential. When demand for NAD(+) exceeds the demand for ATP in cells, aerobic glycolysis is promoted despite available oxygen.

RACK1 mediates the advanced glycation end product-induced degradation of HIF-1α in nucleus pulposus cells via competing with HSP90 for HIF-1α binding

Yi-Chang Xu et al.

Summary: This study assessed the mechanistic relationship between AGE accumulation and HIF-1 alpha degradation in NPCs, revealing that AGE treatment leads to reduced HIF-1 alpha protein levels through RACK1-mediated proteasomal degradation, independently of the canonical PHD-mediated degradation pathway. The competition between RACK1 and HSP90 for HIF-1 alpha binding following AGE treatment suggests potential avenues for the treatment or prevention of intervertebral disc degeneration.

Melatonin Targets Metabolism in Head and Neck Cancer Cells by Regulating Mitochondrial Structure and Function

Ana Guerra-Librero et al.

Summary: Melatonin induces a shift to aerobic mitochondrial metabolism in head and neck cancer cells, resulting in changes in mitochondrial morphology, function, fusion, and fission, increased oxidative phosphorylation, and inhibited glycolysis. This leads to increased ROS production, apoptosis, mitophagy, decreased cell proliferation, and suggests new molecular pathways involved in melatonin's anticancer activity. High doses of melatonin used for its cytotoxic effects may also impact HNSCC cells through melatonin receptors.

Metabolic Anti-Cancer Effects of Melatonin: Clinically Relevant Prospects

Marek Samec et al.

Summary: Metabolic reprogramming is crucial for cancer development, and melatonin plays a significant role in inhibiting tumorigenesis and metastasis by regulating key enzymes and glucose transporters, among other pathways. Melatonin also modulates critical metabolic processes and signaling cascades, highlighting its potential as an innovative approach for cancer prevention and therapy.

Mitochondrial ROS promote mitochondrial dysfunction and inflammation in ischemic acute kidney injury by disrupting TFAM-mediated mtDNA maintenance

Meng Zhao et al.

Summary: Reducing mtROS levels attenuated renal dysfunction, mitochondrial damage, and inflammation in IRI-AKI mice. The loss of TFAM can impact mtDNA maintenance, mitochondrial energy metabolism, and cytokine release. Insights into the role of mtROS and TFAM in IRI-AKI provide potential therapeutic targets for renal repair.
