Solid-state 17O NMR study of α-d-glucose: exploring new frontiers in isotopic labeling, sensitivity enhancement, and NMR crystallography

Summary: This study reports the synthesis and solid-state O-17 NMR characterization of alpha-d-glucose with all six oxygen atoms specifically labeled with O-17. By utilizing paramagnetic Cu(II) doping, new CPMAS CryoProbe technology, and apodization weighted sampling, the sensitivity of solid-state O-17 NMR was significantly enhanced, enabling the acquisition of high-quality 2D O-17 MQMAS spectra for carbohydrate compounds. This work represents the first full characterization of the O-17 NMR properties of all oxygen-containing functional groups in a carbohydrate molecule and aided NMR signal assignments through GIPAW calculations.

17O NMR Studies of Yeast Ubiquitin in Aqueous Solution and in the Solid State

Summary: A general method for amino acid-type specific O-17-labeling of recombinant proteins in Escherichia coli has been reported. This study demonstrates the first detection of O-17 NMR signals from protein terminal residues and side chains in aqueous solution, as well as solid-state O-17 NMR spectra for labeled proteins obtained at an ultrahigh magnetic field. These findings represent a significant advancement in the field of O-17 NMR studies of proteins.

Isotropic solid-state MQMAS NMR spectra for large quadrupolar interactions using satellite-transition selective inversion pulses and low rf fields

Summary: The multiple-quantum magic-angle spinning (MQMAS) pulse sequences allow for obtaining isotropic NMR spectra for large quadrupolar interactions using lower rf fields. By selectively inverting the satellite-transitions with low-power pulses, efficient excitation and conversion of triple-quantum coherences is achieved even for large quadrupolar couplings. The low-power multiple-quantum magic-angle spinning (lpMQMAS) pulse sequences have been demonstrated on samples with large quadrupolar interactions, showing promising results for isotropic NMR spectra.

17O solid-state NMR at ultrahigh magnetic field of 35.2 T: Resolution of inequivalent oxygen sites in different phases of MOF MIL-53(Al)

Summary: In this study, the use of O-17 isotopic enrichment and O-17 SSNMR experiments at an ultrahigh magnetic field of 35.2 T allowed for the complete resolution of all chemically and crystallographically inequivalent oxygen sites in two representative MIL-53(Al) phases. This advancement enhances the potential for using NMR crystallography approach to determine crystal structures of new MOFs and verify existing MOF structures.

Looking into the dynamics of molecular crystals of ibuprofen and terephthalic acid using 17O and 2H nuclear magnetic resonance analyses

Summary: This study investigates the structure and dynamics of crystalline ibuprofen and terephthalic acid using O-17 and H-2 NMR analyses, with improved labelling protocols and various experimental methods. The dynamics around carboxylic groups of ibuprofen were studied, revealing motions related to concerted double proton jump and H-bonded unit flip. Preliminary experiments with a H-2-O-17 diplexer connected to the probe were also conducted, showing potential for recording NMR spectra at different temperatures without probe configuration changes.

Two- and Three-Dimensional 13C-17O Heteronuclear Correlation NMR Spectroscopy for Studying Organic and Biological Solids

The experiments were performed on different samples using various schemes to achieve coherence transfer between C-13 and O-17 nuclei. This new 3D O-17 NMR experiment aims for site-resolved solid-state O-17 NMR studies, demonstrating successful correlations between O-17, C-13, and C-13 nuclei.
