Characterization of a Cryptic NRPS Gene Cluster in Bacillus velezensis FZB42 Reveals a Discrete Oxidase Involved in Multithiazole Biosynthesis

Qiyao Shen et al.

Summary: The biosynthetic mechanism of polythiazole in microbial nonribosomal peptides (NRPs) has been uncovered, revealing the involvement of a cryptic NRPS gene cluster. The study also showed that the oxidation of polythiazole in NRPs is not catalyzed by integrated oxidase domains as previously believed, but by a standalone oxidase.

Complete genome sequence of a marine-derived bacterium Pseudomonas sp. SXM-1 and characterization of its siderophore through antiSMASH analysis and with mass spectroscopic method

Wenbin Guo et al.

Summary: The complete genome of Pseudomonas sp. SXM-1 isolated from the coastal sea water of Xiamen Bay was presented, which will aid in genome mining of the pyoverdine synthetase coding gene cluster. Analysis revealed a circular chromosome with specific GC base content and encoding gene numbers, and the structure of pyoverdine produced by Pseudomonas sp. SXM-1 was predicted and characterized using tools and methods.

Rufomycins or Ilamycins: Naming Clarifications and Definitive Structural Assignments

Bin Zhou et al.

Summary: The paper discusses the structures, naming, and anti-Mycobacterium tuberculosis activity of cyclopeptides such as rufomycin and ilamycin. Through NMR spectroscopy, the configuration and diastereoisomers of these compounds were determined, and semisynthetic derivatives were prepared for validation. Structural differences and conformers were analyzed to understand the biological activity of these compounds, leading to a unified nomenclature for the group.

Global Genome Mining Reveals a Cytochrome P450-Catalyzed Cyclization of Crownlike Cyclodipeptides with Neuroprotective Activity

Jessie James Limlingan Malit et al.

Summary: By conducting global genome mining of bacterial genomes, 829 cyclodipeptide biosynthesis gene clusters containing a cytochrome P450 gene were identified. Heterologous expression of these gene clusters led to the discovery of two novel crownlike cyclodipeptides, cyctetryptomycin A and B, which exhibit complex macrocycle structures and neuroprotective activity. Two cytochrome P450 enzymes catalyze sequential reactions resulting in the cyclization of diketopiperazine dimers within the gene clusters.
