Astaxanthin-Mediated Bacterial Lethality: Evidence from Oxidative Stress Contribution and Molecular Dynamics Simulation

Jamiu Olaseni Aribisala et al.

Summary: The study evaluated the role of oxidative stress in astaxanthin-mediated antibacterial action through both in silico and in vitro analyses. Results suggest that oxidative stress may be involved in the antibacterial mechanism of astaxanthin, as evidenced by the increase in superoxide anion levels in tested organisms following treatment with astaxanthin. The higher free energy scores of astaxanthin's complexes with topoisomerase IV ParC and ParE compared to reference antibiotics indicate a potential role of oxidative stress in the antibacterial capability of astaxanthin.

Design and synthesis of pyrazolone-based compounds as potent blockers of SARS-CoV-2 viral entry into the host cells

Vincent A. Obakachi et al.

Summary: This study aimed to design and synthesize novel pyrazolone-based compounds as potential inhibitors to interrupt the interaction between the viral spike protein and the host cell receptor. Computational analysis showed that certain compounds had higher affinity to SARS-CoV-2 Sgp and hACE-2 compared to standard drugs, suggesting that pyrazolone-based compounds may be potent blockers of viral entry into host cells.

COVID-19 vaccines: concerns beyond protective efficacy and safety

Chih-Cheng Lai et al.

Summary: Four vaccines have been granted emergency use authorization, 87 are in clinical development, and 186 are in pre-clinical development. The development of COVID-19 vaccines is rapidly progressing, but the benefits must outweigh the potential risks. Collaboration between clinicians, healthcare authorities, and manufacturers is essential for successful vaccination programs to control the COVID-19 pandemic.

The interaction of the severe acute respiratory syndrome coronavirus 2 spike protein with drug-inhibited angiotensin converting enzyme 2 studied by molecular dynamics simulation

Babak Nami et al.

Summary: The study suggests that using ACE2 inhibitors may increase the risk of SARS-CoV-2 infection and worsen COVID-19 disease outcomes. Additionally, it was found that SARS-CoV-2 can negate the function of ACE2 inhibitors and restore the enzymatic activity of ACE2. Therefore, ACE2 inhibition is not a useful treatment against COVID-19 infection.

The TMPRSS2 Inhibitor Nafamostat Reduces SARS-CoV-2 Pulmonary Infection in Mouse Models of COVID-19

Kun Li et al.

Summary: The serine protease inhibitors camostat and nafamostat have shown potential in limiting SARS-CoV-2 infection in respiratory tract, with nafamostat exhibiting greater potency. Preclinical evidence supports the efficacy of nafamostat in treating and/or preventing COVID-19 in mice models sensitized to SARS-CoV-2 infection by introducing human ACE2.
