Serpin Signatures in Prion and Alzheimer's Diseases

Summary: This study investigated the differential expression of serpin superfamily members in neurodegenerative diseases. The results showed dysregulation of SERPINB1, SERPINB6, SERPING1, SERPINH1, and SERPINI1 in sCJD individuals compared to controls, while only SERPINB1 was upregulated in AD patients. Additionally, SerpinA3n transcript and protein were upregulated in a mouse model of AD. These findings suggest that SERPINA3/SerpinA3n may be a potential therapeutic target for the treatment of prion and prion-like neurodegenerative diseases.

Pharmacological inactivation of the prion protein by targeting a folding intermediate

Summary: The researchers introduced a new drug discovery approach called Pharmacological Protein Inactivation by Folding Intermediate Targeting (PPI-FIT), which targets folding intermediates of disease-causing proteins. Testing this approach on the cellular prion protein, they identified ligands that could stabilize a folding intermediate and promote its degradation, supporting the idea that modulating protein levels by acting on their folding pathways may have therapeutic potential. Moreover, recent computational advancements in simulating biochemical processes have allowed for investigating protein regulation mechanisms with realistic physics-based models at an atomistic level.

Tackling prion diseases: a review of the patent landscape
