Oncogenic AURKA-enhanced N6-methyladenosine modification increases DROSHA mRNA stability to transactivate STC1 in breast cancer stem-like cells

Fei Peng et al.

Summary: RNase III DROSHA is upregulated in various cancers and interacts with beta-Catenin to activate STC1 in a RNA cleavage-independent manner. The enhanced stability of DROSHA mRNA through m(6)A modification in BCSCs, activated by AURKA, plays a key role in promoting the BCSC phenotype. Inhibition of DROSHA m(6)A modification attenuates BCSC traits, highlighting the importance of AURKA-induced m(6)A modification in breast cancer progression.

High Wilms' tumor 1 associating protein expression predicts poor prognosis in acute myeloid leukemia and regulates m6A methylation of MYC mRNA

Duolan Naren et al.

Summary: This study found that high expression of WTAP in AML patients is associated with poor prognosis, and WTAP plays an epigenetic role in AML by regulating cell proliferation, tumorigenesis, cell cycle, and differentiation. The research also indicated that WTAP makes AML cells resistant to daunorubicin.

m6A-RNA Demethylase FTO Inhibitors Impair Self-Renewal in Glioblastoma Stem Cells

Sarah Huff et al.

Summary: N-6-methyladenosine (m(6)A) is the most abundant mRNA modification that regulates gene expression and plays a key role in various diseases. The FTO inhibitor FTO-04 has been shown to effectively inhibit neurosphere formation in glioblastoma stem cells, making it a potential new lead for treating glioblastoma.

Summary: BRAF and KRAS mutations play a significant role in NSCLC by activating the RAS/RAF/MEK/ERK signaling pathway. Current approved therapies include BRAF and MEK inhibitors, while combination with other drugs may enhance therapeutic effects. Further research is needed to explore optimal drug combinations for treating lung cancer patients.

A novel hypoxic long noncoding RNA KB-1980E6.3 maintains breast cancer stem cell stemness via interacting with IGF2BP1 to facilitate c-Myc mRNA stability

Pengpeng Zhu et al.

Summary: In a hypoxic microenvironment, a long noncoding RNA called KB-1980E6.3 was found to be upregulated in breast cancer tissues, promoting self-renewal of breast cancer stem cells and tumorigenesis. It exerts its function through the lncRNA KB-1980E6.3/IGF2BP1/c-Myc signaling axis, maintaining stemness.

Summary: Adipose-derived stem cells have low immunogenicity and various self-renewal abilities, can differentiate into different tissue-specific progenitors, migrate into damaged sites, and act through autocrine and paracrine pathways. However, the lack of standardization protocols and discrepancies in cell outcomes and engraftment pose limitations on their widespread use.

Small-molecule inhibition of METTL3 as a strategy against myeloid leukaemia

Eliza Yankova et al.

Summary: The study identified a highly potent and selective catalytic inhibitor of METTL3, STM2457, which shows potential therapeutic value in treating AML. Treatment with this inhibitor leads to reduced AML growth, increased differentiation and apoptosis, and decreased m(6)A levels on leukemogenic mRNAs. In vivo pharmacological inhibition of METTL3 results in impaired engraftment and prolonged survival in mouse models of AML.

FTO-mediated cytoplasmic m6Am demethylation adjusts stem-like properties in colorectal cancer cell

Sebastien Relier et al.

Summary: The demethylase FTO was shown to remove N6-methyladenosine (m6A) and N6, 2'-O-dimethyladenosine (m6A(m)) modifications on RNAs. Here the authors show that FTO impedes cancer stem cell-like abilities in colorectal cancer cells through its m6A(m) demethylase activity, not through internal m6A demethylase activity.
