Allovalency observed by transferred NOE: interactions of sulfated tyrosine residues in the N-terminal segment of CCR5 with the CCL5 chemokine

Summary: Sulfation of tyrosine residues Y14 and Y15 in the N-terminal segment of CCR5 is essential for high-affinity binding to CCL5, with disulfation resulting in up to a threefold increase in affinity. The sulfated tyrosine residues interact with the same binding pocket on CCL5 and can individually occupy the site with varying affinity, suggesting the presence of allovalency in sulfated Nt-CCR5 peptides. This allovalency, along with stronger electrostatic interactions, may contribute to the increased affinity of disulfated peptides for CCL5.

Summary: Protein tyrosine O-sulfation is crucial for various biological processes, but sulfated tyrosine residues are challenging to detect due to their instability and low abundance. This study engineered SH2 mutants as an antisulfotyrosine antibody mimic, demonstrating high affinity and specificity to sulfotyrosine through tailored selection schemes. Molecular docking simulations supported the characteristics of these SH2 mutants, enhancing the detection and enrichment of sulfoproteins.

Synthesis and evaluation of peptidic thrombin inhibitors bearing acid-stable sulfotyrosine analogues

Sulfotyrosine-Mediated Recognition of Human Thrombin by a Tsetse Fly Anticoagulant Mimics Physiological Substrates

Summary: The tsetse thrombin inhibitor (TTI) is a potent anticoagulant with sub-picomolar inhibitory activity against thrombin, and its activity is significantly enhanced by tyrosine O-sulfate post-translational modification. TTI's unique mode of inhibition, coupled with its high efficacy and small size, makes it an attractive scaffold for designing novel antithrombotics. Through a combination of chemical synthesis and X-ray crystallography, the mechanism of TTI's inhibitory activity against thrombin was revealed.
