Fast Kinetics Reveals Rate-Limiting Oxidation and the Role of the Aromatic Cage in the Mechanism of the Nicotine-Degrading Enzyme NicA2

Margarita A. Tararina et al.

Summary: The research elucidated the mechanism of NicA2, showing its preference for oxidizing tertiary amines with an efficient reductive half-reaction and very slow oxidative half-reaction, suggesting that the true oxidizing agent is unknown.

A cytochrome c is the natural electron acceptor for nicotine oxidoreductase

Mark Dulchavsky et al.

Summary: Nicotine oxidoreductase NicA2 shows potential therapeutic value in aiding smoking cessation by oxidizing nicotine into a non-psychoactive metabolite, although its O-2-dependent activity is poor. Surprisingly, NicA2 functions as a dehydrogenase by donating electrons to cytochrome c instead of using O-2 as an oxidant, challenging previous assumptions about enzymes in the flavin-containing amine oxidase family. This discovery opens up new perspectives for engineering NicA2 as a potentially useful therapeutic agent.
