Myocardial Rev-erb-Mediated Diurnal Metabolic Rhythm and Obesity Paradox

Shiyang Song et al.

Summary: This study investigates the function of cardiac Rev-erb in heart diseases and the relationship between cardiac molecular clock dysfunction and the progression of naturally occurring human heart diseases. The researchers found that Rev-erb knockout in mice leads to dilated cardiomyopathy and heart failure. They also discovered the role of cardiac molecular clock in the obesity paradox and found altered temporal patterns of cardiac Rev-erb gene expression in human hearts with dilated cardiomyopathy. The findings suggest that targeting myocardial bioenergetics downstream of Rev-erb may be a potential approach for treating heart failure and dilated cardiomyopathy.

NF-κB modifies the mammalian circadian clock through interaction with the core clock protein BMAL1

Yang Shen et al.

Summary: Recent studies have shown that NF-kappa B plays a significant role in directly regulating the circadian clock, altering clock oscillations in both cells and tissues, as well as in mice. NF-kappa B competes with coregulators like CRY1 and CBP/p300 for BMAL1 binding to affect circadian transcription, linking inflammation to circadian timekeeping.

Mitochondrial autophagy and cell survival is regulated by the circadian Clock gene in cardiac myocytes during ischemic stress

Inna Rabinovich-Nikitin et al.

Summary: This study reveals that the CLOCK gene transcriptionally coordinates mitochondrial quality control mechanisms in cardiac myocytes, regulating an adaptive stress response crucial for cell survival. Restoring CLOCK activity may be beneficial in reducing cardiac injury in individuals with disrupted circadian rhythms.

The Impact of Melatonin Supplementation and NLRP3 Inflammasome Deletion on Age-Accompanied Cardiac Damage

Ramy K. A. Sayed et al.

Summary: The study investigated the role of NLRP3 inflammasome in cardiac aging by evaluating age-related changes in cardiac muscles and the effects of melatonin therapy. The results showed that cardiac sarcopenia begins at 12 months, with significant changes in cardiac myocytes and mitochondria, while NLRP3(-/-) mice exhibited fewer age-related changes. Melatonin therapy showed beneficial effects in preserving cardiomyocyte structure and reducing cardiac hypertrophy.
