Recent advances in mass spectrometry analysis of neuropeptides

Summary: Neuropeptides have become the focus of many recent research studies due to their involvement in numerous biochemical pathways. Traditional analytical methods are limited in terms of global investigations, leading researchers to search for more advanced techniques such as mass spectrometry (MS). MS has provided a methodology to gain global knowledge of a neuropeptidome on a spatial, temporal, and quantitative level.

In-Depth Site-Specific O-Glycosylation Analysis of Glycoproteins and Endogenous Peptides in Cerebrospinal Fluid (CSF) from Healthy Individuals, Mild Cognitive Impairment (MCI), and Alzheimer's Disease (AD) Patients

Summary: Site-specific O-glycoproteome mapping in complex biological systems provides a molecular basis for understanding the structure-function relationships of glycoproteins and their roles in physiological and pathological processes. A comprehensive strategy was developed to analyze the O-glycoproteome in cerebrospinal fluid (CSF) and identify O-glycopeptides from both sialylated and nonsialylated glycoforms. The study also revealed that endogenous peptides in CSF were predominantly O-glycosylated, with a decreasing trend in fucosylation and an increasing trend in endogenous peptide O-glycosylation observed in patients with Alzheimer's disease.

A genetically encoded sensor for in vivo imaging of orexin neuropeptides

Summary: This study developed a genetically encoded orexin sensor called OxLight1, which enables sensitive and direct optical detection of orexin neuropeptides in living animals with high spatiotemporal resolution. Bioinformatic analysis of peptidomics data sets should include specific post-translational modifications critical for the function of bioactive peptides.

The plasma peptides of Alzheimer's disease

Summary: Comparing endogenous tryptic peptides from Alzheimer's patients and controls, proteins specific to Alzheimer's dementia were discovered with higher observation frequency and intensity in the plasma of Alzheimer's patients. These proteins included common plasma proteins like complement C2, C7, and C1QBP, as well as specific cellular gene symbols and phosphopeptides associated with Alzheimer's. STRING analysis revealed an increase in cytoplasmic proteins and proteins involved in various cellular functions in Alzheimer's plasma.
