Summary: The human monoclonal antibody C10 exhibits extraordinary cross-reactivity, potently neutralizing Zika virus (ZIKV) and the four serotypes of dengue virus (DENV1-DENV4). The C10 Fab has high affinity for ZIKV and DENV1 but not for DENV2, DENV3, and DENV4, due to the limited binding landscape of the E protein conformation. However, the icosahedral organization of the viruses allows two different dimers to be reached by both Fab arms of a C10 immunoglobulin, which contributes to its exceptional neutralization breadth.

Recent studies show that besides viral structural proteins, nonstructural proteins also play key roles in coordinating assembly and packaging of viral genome. This review highlights recent advances in understanding flavivirus virion assembly, including molecular interactions between viral structural and nonstructural proteins, host factors, and features of viral genomic RNA necessary for efficient assembly.
