Separation of phosphorothioated oligonucleotide diastereomers using multiplexed drift tube ion mobility mass spectrometry

Summary: Hydrophilic interaction liquid chromatography coupled to drift tube ion mobility spectrometry was used to separate diastereomers of five-unit oligonucleotides with different phosphorothioate linkages. The study investigated multiplexed drift tube ion mobility and post-acquisition processing using a demultiplexing tool. By optimizing the electric field inside the drift tube, the diastereomers could be separated unambiguously. The use of multiplexed ion mobility and a high-resolution demultiplexing tool represents a breakthrough in resolving diastereomers in linear drift tube ion mobility spectrometry.

Summary: Marfey's reagent, introduced in 1984, has been the most successful method for enantioseparation of amino acids and amino group containing compounds through pre-column derivatization in high-performance liquid chromatography. By substituting the F atom with different chiral auxiliaries, multiple chiral variants can be prepared to tailor the hydrophobicity and resolution of diastereomeric derivatives. Various applications of Marfey's reagent and its chiral variants for enantioseparation of certain amino group containing drugs/amino acids have been assessed in this study.

The method allows for rapid chiral analysis within 3 minutes per sample and is robust in samples containing high amounts of salts.
