T cell apoptosis characterizes severe Covid-19 disease

Sonia Andre et al.

Summary: By comparing COVID-19 patients with healthy donors, the study found a strong correlation between T cell apoptosis and lymphopenia in severe cases. The plasma levels of soluble FasL (sFasL) and T cell surface expression of Fas/CD95 were positively correlated with T cell death and CD4 T cell counts. The study also identified modulations in the Bcl-2 family and demonstrated the potential of a pan-caspase inhibitor, Q-VD, to prevent T cell apoptosis and enhance Th1 transcripts.

Summary: This study found that the H7N9 virus can replicate efficiently in mouse lungs and activate gasdermin E (GSDME)-mediated pyroptosis, leading to a cytokine storm. Knockout of the Gsdme gene changed the death pathway of alveolar epithelial cells from pyroptosis to apoptosis, and Gsdme knockout mice survived lethal H7N9 virus infection. The Epstein-Barr Virus Oncogene EBNA1 Suppresses Natural Killer Cell Responses and Apoptosis Early after Infection of Peripheral B Cells

Danielle Westhoff Smith et al.

Summary: NK cells are an essential part of the innate immune system, able to directly kill cells and secrete cytokines. Epstein-Barr virus (EBV) can suppress NK cell responses and cell death in newly infected cells by downregulating NKG2D ligands and modulating c-Myc expression through its EBNA1 oncogene. This allows infected cells to evade immune surveillance and apoptosis, contributing to lifelong latency of the virus in memory B cells.

This study found that human astrocytes infected with ZIKV underwent necroptosis, as evidenced by increased phosphorylation of RIPK1, RIPK3, and MLKL proteins. Inhibition of RIPK3, but not RIPK1, suppressed cell death, indicating RIPK1-independence in ZIKV-induced necroptosis.
