Summary: This study evaluated the expression of AR, ER, PR, and HER2 in UC/DEAC and other subtypes of high-grade endometrial carcinomas. The findings showed high expression of AR in UC/DEAC, as well as in the majority of FIGO 3 endometrioid carcinomas, serous carcinomas, and carcinosarcoma.

Signal transduction pathway activity in high-grade serous carcinoma, its precursors and Fallopian tube epithelium

Phyllis van der Ploeg et al.

Summary: This study investigated the activity of key signal transduction pathways in serous tubal intraepithelial carcinoma (STIC) and high-grade serous carcinoma (HGSC) and compared them to normal Fallopian tube epithelium (FTE). The results showed that the ER pathway activity was lower, while the PI3K and HH pathway activity was higher in STIC and HGSC compared to FTE. This suggests that these pathways may play a crucial role in the neoplastic transformation of FTE into STIC.

Cancer of the corpus uteri: 2021 update

Summary: Endometrial cancer is the most common gynecological malignancy in high- and middle-income countries, with a tendency for high-grade cancers to recur. Tailoring treatment to tumor biology is crucial for balancing efficacy and toxicity, with molecular factors increasingly used for defining prognosis and treatment. Standard treatment includes surgery and adjuvant therapy based on risk factors, with chemotherapy particularly used in non-endometrioid cancers and those with TP53 mutation.

Summary: The study demonstrates the potent antiproliferative, proapoptotic, anti-migration, and anti-invasive properties of the newly synthesized androstane derivative 17APAD in cervical cancer cell lines, independent of androgenic activity. These findings suggest that 17APAD may be a promising prototype for designing novel anticancer agents and warrant further research in this area.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

TLR4/IL-6/IRF1 signaling regulates androgen receptor expression: A potential therapeutic target to overcome taxol resistance in ovarian cancer

Summary: The study reveals that TLR4 and IL-6 play a crucial role in chemoresistance in ovarian cancer by regulating AR gene expression. These proteins interact in signaling pathways, affecting AKT activation and IRF1 regulation. Clinical findings suggest that high IL-6 expression may lead to decreased progression-free survival in taxol-treated ovarian cancer patients.

Androgen Receptor-Dependent Mechanisms Mediating Drug Resistance in Prostate Cancer

Marzieh Ehsani et al.

Summary: Prostate cancer can develop resistance to hormone treatment and chemotherapy, mainly due to the androgen receptor (AR). There are genomic and non-genomic signaling bypass mechanisms that tumor cells may develop to escape therapy. As resistance progresses, tumors may evolve further adaptive pathways which lead to increased aggressiveness and metastatic ability.

Summary: Higher circulating concentrations of total and free testosterone were associated with higher endometrial cancer risk, while an inverse association was found for SHBG. Genetically predicted IGF-1 concentration was not associated with endometrial cancer risk. Our findings suggest a probable causal relationship between testosterone and SHBG with endometrial cancer risk.

Summary: Ovarian cancer primarily originates from three cellular origins, with over 85% being epithelial ovarian carcinomas, and cancer stem/progenitor cells are considered cancer promoters. Activation of the androgen receptor is linked to the development and progression of ovarian cancer, with experimental studies showing positive effects of anti-androgen or anti-AR reagents in suppressing disease progression.
