Targeting p53 for the treatment of cancer

Michael J. Duffy et al.

Summary: Dysfunction of the TP53 gene is prevalent in human malignancies, making it an attractive target for new anticancer drugs. Two strategies, reactivating mutant p53 and inhibiting the interaction between wild-type p53 and MDM2/MDM4, are being explored in clinical trials. The development of compounds targeting these strategies has the potential to revolutionize cancer treatment.

The purple mangosteen (Garcinia mangostana): Defining the anticancer potential of selected xanthones

Mirielle C. Nauman et al.

Summary: The purple mangosteen is a popular fruit in Southeast Asia that contains xanthones, which have significant anti-cancer and anti-tumor activities. Alpha-mangostin, the most abundant xanthone, has been extensively studied for its anticancer effects, while other xanthones and mangosteen extracts also show promise but their mechanisms of action are not well understood. Clinical anticancer activity of mangosteen products has not been evaluated.

Expression of TIMPs and MMPs in Ovarian Tumors, Ascites, Ascites-Derived Cells, and Cancer Cell Lines: Characteristic Modulatory Response Before and After Chemotherapy Treatment

Ruth M. Escalona et al.

Summary: This study analyzed the expression of TIMPs and associated MMPs in different stages and grades of serous ovarian tumors, as well as their response to chemotherapy treatment. The results showed that TIMPs and MMPs are differentially expressed in ovarian tumors and are associated with cancer stem cells.

The Effect of α-Mangostin and Cisplatin on Ovarian Cancer Cells and the Microenvironment

Paulina Borzdzilowska et al.

Summary: This study evaluated the effects of cisplatin and a plant extract alpha-mangostin on ovarian cancer cells and the cancer cell microenvironment. The results showed that the combination of alpha-mangostin and cisplatin reduced the concentration of cisplatin while maintaining the same cytotoxic effect. Additionally, the composition of exosomes released from drug-treated cancer cells may be different from untreated cells.

Adenomatous polyposis coli in cancer and therapeutic implications

Olivia Noe et al.

Summary: Inactivating mutations of the APC gene and upregulation of the Wnt signaling pathway play crucial roles in colorectal cancer development, with potential implications in other cancers as well. Research for therapies targeting APC mutations is ongoing, with a focus on identifying effective molecular targets to improve patient outcomes.

Summary: Every year, the American Cancer Society projects the numbers of new cancer cases and deaths in the United States, with the latest data showing a significant decline in lung cancer mortality, while prostate cancer mortality has plateaued and breast and colorectal cancer mortality have slowed. Improvements in treatment have accelerated progress against lung cancer, leading to a record drop in overall cancer mortality.

Exosome-mediated transfer of CD44 from high-metastatic ovarian cancer cells promotes migration and invasion of low-metastatic ovarian cancer cells

Xiameng Shen et al.

Summary: Tumor-derived exosomes play important roles in promoting migration and invasion of ovarian cancer cells. CD44, transferred through exosomes, influences the metastatic phenotype of cancer cells and may be a potential therapeutic target for ovarian cancer.

CD44: A Multifunctional Mediator of Cancer Progression

Malak Hassn Mesrati et al.

Summary: CD44, a non-kinase cell surface transmembrane glycoprotein, is widely implicated as a cancer stem cell marker in multiple cancers. Cells overexpressing CD44 possess CSC traits and resistance to therapy. The CD44 gene undergoes alternative splicing, resulting in different isoforms that interact with various ligands and drive cancer-associated signaling pathways. However, the association between CD44 expression levels and clinicopathological features is contradictory, although high CD44 expression significantly contributes to tumorigenic mechanisms.

Adenomatous Polyposis Coli loss controls cell cycle regulators and response to paclitaxel in MDA-MB-157 metaplastic breast cancer cells

Emily M. Astarita et al.

Summary: APC loss is associated with 70% of sporadic breast cancers, especially in TNBC. Knockdown of APC in TNBC cells induced paclitaxel resistance, with studies showing alterations in cell cycle-related genes and proteins, suggesting potential for combination therapy to resensitize APC-mutant breast cancers to taxanes.
